摘　　要：目的:观察选择性JAK/STAT3抑制剂JSI-124对舌鳞状细胞癌Tca-8113细胞增殖抑制以及胞质转录因子STAT3蛋白表达的影响。方法:用MTT、流式细胞仪分别检测Tca-8113细胞在不同浓度JSI-124及不同时间作用下,细胞增殖及凋亡的情况;Western Blot法检测作用后细胞中STAT3蛋白及其磷酸化蛋白表达的变化,并对所得结果进行统计分析。结果:JSI-124可以抑制舌鳞癌Tca-8113细胞的增殖,其抑制作用随着时间的延长和药物浓度的增加而增强;STAT3蛋白的表达无明显变化,而磷酸化激活状态的STAT3蛋白表达显著降低(P<0.01)。结论:选择性JAK/STAT3抑制剂JSI-124可明显抑制舌鳞癌Tca-8113细胞的增殖并诱导其细胞凋亡。Objective： To study the effect of JSI-124 on the proliferation of tongue squamous cell carcinoma Tca-8113 cells.Methods： MTT assay and flow cytometry were used to examine the proliferation and apoptosis of Tca-8113 cells after treatment with JSI-124 at different concentrations and for different times respectively.Western Blot was employed to examine STAT3 and p-STAT3 protein expression in the treated cells.Results： JSI-124 suppressed the cell proliferation and induced the cell apoptosis in a dose and time dependant manner.JSI-124 did not change STAT3 protein expression,but decreased p-STAT3 protein expression（P0.01）.Conclusion： JSI-124 can inhibit proliferation and induce apoptosis of Tca-8113 cells.
