珍菊降压片中药物配伍对大鼠肾脏表达谱的影响

摘　　要：目的研究珍菊降压片及组方中中药与西药部分对大鼠肾脏表达谱的影响,探讨其中西药物配伍的合理性。方法将24只Wistar大鼠随机均分成空白对照组、珍菊降压片复方低、高剂量组(200、800 mg.kg-1)、相应剂量西药组(4.4、17.8mg.kg-1)、复方高剂量相应剂量中药组(782 mg.kg-1)。连续灌胃给药13周后,分离肾脏,抽提、纯化总RNA,芯片杂交后进行扫描分析。结果与空白对照组相比,基因表达谱的改变顺序为复方低剂量组<中药高剂量组<西药低剂量组<复方高剂量组<西药高剂量组,复方低剂量与西药低剂量组的基因聚类结果显著分开,接近于空白对照组,复方高剂量组与西药高剂量组的基因聚类结果有一定比例的改变,但尚不能完全分开,各组间差异主要表现在离子通道、凋亡、信息传导途径等相关基因群的表达改变不同。结论珍菊降压片影响肾脏基因表达改变主要源于西药部分,中药配伍后可降低西药部分所引起的基因表达水平的改变,显示其一定的减毒作用。OBJECTIVE To study the effect of the combination of traditional Chinese medicine and chemical drugs in Zhenju Jiangya tablets on the gene expression spectrum in rat kidney.METHODS 24 Wister rats were randomly divided into six groups,blank control group,the low and high dose group of Zhenju Jiangya tablet（200 mg·kg-1 and 800 mg·kg-1）,the corresponding dose group of the western medicine group 4.4 mg·kg-1 and 17.8 mg·kg-1,the corresponding high dose group of the traditional Chinese medicine group（782 mg·kg-1）,with four rats in each group.Kidneys were separated from the rats after po administration for thirteen weeks and the change of kidney gene expression spectrum was tested with gene chip technology.RESULTS The gene expression levels in all groups were changed.There were some differences presented in the following aspects：ion channel,cell apoptosis,and signal transduction pathway.CONCLUSION The effect of Zhenju Jiangya tablets,which contains both traditional Chinese medicines and chemical drugs,is developed not only by the two groups separately,but also by the synergistic effect of the groups.The addition of traditional Chinese medicine,to a certain extent,decreased the changes of gene expression induced by chemical drugs.
