摘 要:目的 在体外建立神经组织生长趋化性模型,探讨神经生长因子(NGF)对神经组织的趋化作用。 方法 直径约1mm的玻璃毛细管吸取约10μl的染料溶液,观察染料在培养液中的扩散;同样毛细管吸取含NGF的溶液与鸡胚神经节共培养,观察神经组织的生长方向。 结果 直径1mm的毛细管可稳定吸取10μl的液体;吸取的染料在毛细管周围形成分布均匀的浓度梯度;培养的神经节组织向高NGF浓度梯度方向生长。 结论 毛细管模型是研究神经组织趋化性生长的良好的模型;NGF对神经组织生长具有趋化作用。Objectives The purpose of this study is to found a reliable model of neurotropism in vitro for characterizing neurotropic factors,and to evaluate the neurotropic effect of NGF on the cultured nerve tissue Methods (1)Ten capillaries of about 1 mm in diameter containing a red dye were put down to the culture media in dishes through the dish cover Then the diffusion patterns of the dye around the lower end of the capillaries were observed (2)Eight DRGs were cultured in 8 dishes with capillary containing NGF solution The other 8 DRGs were cultured in dishes with capillaries containing plain culture media as the control Results One piece of capillary with a diameter of 1~1 5 cm could safely absorb 10 μl of fluid While the capillary containing ink was put down to the bottom of the culture dish coated with a piece of nylon pad,the red color slowly disseminated with heavy staining in the center and light staining in the peripheral portion This pattern of shade could simulate the concentration gradient of trophic factors in this model There were significantly more nerve fibres growing from the DRGs towards the capillary in the experimental group than in the control group Conclusion The capillary model was effective in studying the chemotactic growth of nerve tissue NGF had definite neurotropic effects on DRGs tissue
