摘 要:目的:研究TRPC通道在难治性颞叶癫痫(RTLE)患者脑组织中的表达。方法:收集27例RTLE患者为研究组,8例与其年龄、性别相匹配的颅脑手术减压或清创患者为对照组,取颞叶组织为研究部位。采用HE染色确认解剖部位及病理类型,免疫组化方法检测TRPC通道蛋白的表达。结果:研究组颞叶胶质细胞呈不同程度增生,部分有噬神经现象和神经元变性。研究组颞叶中TRPC1、3、4、5阳性细胞表达较对照组显著增多,差异具有统计学意义(P<0.05或0.01),而TRPC6在2组颞叶均未见表达。结论:TRPC1、3、4、5通道蛋白在RTLE患者颞叶组织中表达上调,提示TRPC通道可能与RTLE的发病有关。Objective: To investigate expression of eannonica[ transient receptor potential channels (TRPC channels) in brain tissues from patients with refractory temporal lobe epilepsy (RTLE). Methods: Twenty-seven RTLE patients and 8 age- and gender-matched controls expe- riencing cerebra[ operations for decompression or debridement were recruited for this study. Temporal lobe section was obtained from each individuals. HE staining was used to determine an- atomic site and pathological changes. Immunohistochemistry staining was used to examine the ex- pression of TRPC protein. Results. Gliosis was observed at different levels in the brain tissues from the patients. Neuronophagia and neuronal degeneration were found in some brain tissues from the patients. The expressions of TRPC1,3,4 and 5 protein were significantly up-regulated in the temporal lobe in the patients than those in the controls (P〈0.05 or 0.01). However, TR- PC6 protein was not detected in the 2 group. Conclusion: The expression of TRPC1,3,4 and 5 were up-regulated in the temporal lobes of RTLE patients, suggesting the contribution to epileptogenesis.
