摘 要:目的探讨急性热应激对大鼠节律基因Per2mRNA昼夜表达水平的影响。方法选用雄性Wistar大鼠,随机分为对照组和热暴露组。对照组:置于室温24℃环境中;热暴露组:环境温度为32℃,经7 d热暴露后,采用体温遥感技术监测大鼠体温变化,测定大鼠脑垂体与肾上腺的相对质量及肝脏Per2 mRNA昼夜的表达水平。结果急性热应激对大鼠体温和体质量无影响,但可增加大鼠脑垂体和肾上腺的相对质量,并可显著抑制节律基因Per2 mRNA在夜间的表达水平。结论急性热应激可干扰Per2 mRAN的昼夜节律性表达。Objective To investigate the effect of acute heat stress on the day-night circadian gene Per2 mRNA expression in the liver of rats.Method Male Wistar rats were randomly divided into two groups and exposed to heat at 32℃or to a room temperature at 24℃(control).After 7 days of heat exposure,the body temperature was measured by telemetry.The relative weight of the pituitary and adrenal glands was determined after the exposure,and liver Per2 mRNA expression level was detected using RT-PCR.Results Acute heat stress did not obviously affect body temperature or body weight of the rats.Seven days of heat exposure increased the relative weight of the pituitary and adrenal glands and significantly lowered Per2 mRNA expression level at night.Conclusion Acute heat stress can interfere with the day-night circadian gene Per2 mRNA expression in rats.
