摘  要:目的:建立一种反向高效液相色谱(RP-HPLC)法测定黄连中盐酸小檗碱的含量。方法:采用HPLC法,色谱柱为Luna C18柱(150 mm×4.60 mm,5μm),流动相为乙腈-0.1%磷酸(25∶75),每1 000 ml0.1%的磷酸溶液中加入50μl的三乙胺(扫尾剂)进行洗脱;检测波长为340 nm。结果:采用HPLC法检测黄连中盐酸小檗碱的含量,其在0.246~2.460μg范围内与峰面积积分值呈良好的线性关系。平均加样回收率为100.1%,相对标准偏差(RSD)为1.60%。结论:HPLC简便可行、重复性好,可作为检测盐酸小檗碱含量的质量控制;一定量的三乙胺分析纯可作为很好的扫尾剂,用于盐酸小檗碱的含量测定。Objective: To establish a new RP-high-performance liquid chromatography(HPLC) method for detecting the content of Berberine hydrochloride(BH) in coptis chinensis franch.Methods: The content of BH in coptis chinensis franch was deteminated by HPLC.HPLC conditions for the column: Luna C18(150 mm×4.60 mm,5 μm),acetonitrile-0.1% phosphoric acid(25 ∶75),and adding 50 μl Triethylamine(tailing-suppressing reagent) into every 1 000 ml 0.1% phosphoric acid for elution,the detection wavelength was 340 nm.Results: This HPLC method exhibited a linear relationship between the content of BH within 0.246 to 2.460 μg and the peak area integration,the average recovery rate of BH was 100.1%,the relative standard deviation(RSD) was 1.60%.Conclusion: This is a simple,reproducible and reliable HPLC method for determinating the content of BH.A certain amount of Triethylamine analytically pure reagent is a very good tailing-suppressing reagent for using HPLC method to determinate the content of BH.
