半乳糖凝集素-3在大鼠肝纤维化组织中的表达及意义

摘　　要：目的研究半乳糖凝集素-3(galectin-3,Gal-3)、转化生长因子β1(transforming growth factor beta-1,TGF-β1)、a-平滑肌肌动蛋白(a-smooth muscle actin,a-SMA)在肝纤维化大鼠模型中的动态表达及意义。方法雄性SD大鼠随机分为健康对照组6只,肝纤维化模型组24只,采用二甲基亚硝胺(DMN)腹腔注射制备肝纤维化模型。分别在造模后2、4、6、8周门静脉采血检测血清ALT、AST、Alb,采用HE及Masson染色观察肝组织学变化,反转录聚合酶链反应(RT-PCR)检测不同时间点肝脏galectin-3 mRNA的表达。结果正常肝组织中有galectin-3低表达,2周即升高,4周达高峰,6周下降,差异有统计学意义(P<0.05)。相关性分析显示,galectin-3水平与TGF-β1、a-SMA呈正相关(r=0.882～0.963,P<0.01)。结论 Galectin-3可能与肝纤维化的发生发展过程密切相关。To explore the expression and significance of galectin - 3 （ Gal - 3 ） ,transforming growth factor beta - 1 （TGF - β1 ） and a - smooth muscle actin （ a - SMA） in rat models of hepatic fibrosis. Methods The male SD rats were divided into two groups randomly. The rats were injected with dimethylnitrosamine（DMN） intra -peritoneally to establish the hepatic fibrosis model. The sera were collected from portal vein of the rats at 2, 4, 6,8 weeks after injection. The Histopathologic changes of the liver were observed with HE and MASSON stain. The galectin - 3 mRNA of liver tissues were determined by RT - PCR. Results Galectin - 3 mRNA ex- pression in the liver of normal rats,but the expressions galectin - 3 in fibrosis model group increased gradually with prolonged liver injury than control group, and peaked at 4 weeks and decreased at 6 weeks after injection, which was significantly different at 2,4,6,8 weeks after injections （P 〈 0.05）. And correlation analysis showed that the expression of galectin - 3 mRNA was positively correlated with TGF - 13t and a - SMA （ r = 0. 882 - 0. 963, P 〈 0.01 ）. Conclusion Galectin - 3 may be closely related to the development of liver fibrosis.
