摘　　要：目的应用SHSY5Y神经母细胞瘤建立脑干肿瘤裸鼠模型。方法将SHSY5Y神经母细胞瘤细胞注射到裸鼠脑桥,2.5周后行MRI扫描观察肿瘤生长情况,行苏木精-伊红染色、免疫组化染色观察肿瘤细胞形态及胶质纤维酸性蛋白(GFAP)、巢蛋白(Nestin)和神经蛋白丝(NF)的表达。结果 MRI检查显示脑干部位出现肿瘤,为长Tl、长T2信号,注射钆喷酸葡胺(Gd-DTPA)后信号明显增强。苏木精-伊红染色显示肿瘤为神经母细胞瘤,呈浸润性生长,细胞围绕毛细血管排列成假菊形团结构。免疫组化显示GFAP及NF呈阴性表达,Nestin呈阳性表达。结论 SHSY5Y神经母细胞瘤可用于建立脑干肿瘤动物模型,该模型具有稳定的重复性和可预测性。Objective To establish brainstem tumor model of nude mouse by using SHSY5Y neuroblastoma cells.Methods SHSY5Y neuroblastoma cells were injected into the pons of nude mouse.The growth of tumors was examined by MRI 2.5 weeks after implantation.The morphological characteristics were observed by hematoxylin-eosin staining.The expressions of glial fibrillary acidic protein（GFAP）,Nestin and neurofilament（NF） were detected by immunohistochemistry.Results Tumors were observed in the brainstem by MRI scan with long T1 and long T2 signals,and were enhanced obviously after the injection of Gd-DTPA.The hematoxylin-eosin staining showed that the tumor was neuroblastoma with invasive growth and tumor cells arranging around the capillaries as pseudorosette structure.The immunohistochemistry results showed that tumor cells were negative for GFAP and NF,and postive for Nestin.Conclusions SHSY5Y neuroblastoma cells are used to establish brainstem tumor model of nude mouse,which has stable repeatability and predictability.
