摘 要:目的:探讨抑郁症模型大鼠海马受体、离子通道基因表达谱变化及甘麦大枣汤加味的调节作用。方法:以孤养加慢性不可预见性温和刺激制作抑郁症模型大鼠,通过旷场行为测定和饮水试验观察大鼠行为学变化,采用基因芯片技术检测大鼠海马各类受体mRNA表达,RT-PCR法检测大鼠海马脑源性神经营养因子(BDNF)mRNA表达。并以文拉法辛为对照,观察甘麦大枣汤加味对上述各项指标的影响。结果:模型组大鼠水平运动、垂直活动及糖水消耗量显著下降(P<0.01),海马单胺类(DA、5-HT、NE)、氨基酸类(NMDA、GABA、Gly)、神经肽类(AT、AVP)受体亚型表达下调,BDNF mRNA表达显著下降(P<0.05);与模型组比较,甘麦大枣汤加味23g/kg与文拉法辛25mg/kg大鼠水平运动、垂直活动及糖水消耗量显著上升(P<0.01),海马单胺类(DA、5-HT、NE)、氨基酸类(NMDA、GABA、Gly)、神经肽类(AT、AVP)受体亚型表达上调,BDNF mRNA表达显著增加(P<0.01)。结论:甘麦大枣汤加味与文拉法辛可上调海马单胺类、氨基酸类、神经肽类受体亚型,改善细胞信号转导途径功能,增加BDNF表达,从而纠正模型大鼠行为学异常。Objective:To study changes of gene expression profile of receptors ion channels in hippocampus of rat model with depression and adjusting effects of supplemented Ganmai Dazao Decoction.Methods:A depression rat model was produced by separation and chronic unpredictable mild stress,behaviors of rats were detected by open-field test and sucrose consumption test,mRNA expressions of receptors were detected by technology of gene chip,mRNA expressions of BDNF were detected by RT-PCR.Venlafaxine(25mg/kg) as a control,effects of supplemented Ganmai Dazao Decoctions(crude drug 23g/kg) on datum above-mentioned were observed.Results:Scores of ambulation,rearing and sucrose consumption lowered remarkably(P0.01),mRNA expressions of receptor subtypes of monoamine(DA,5-HT,NE),amino acid(NMDA,GABA,Gly) and neuropeptide(AT,AVP)lowered,mRNA expressions of BDNF lowered remarkably(P0.05) in model group.Comparing with model group,scores of ambulation,rearing and sucrose consumption increased remarkably(P0.01),mRNA expressions of receptor subtypes of monoamine(DA,5-HT,NE),amino acid(NMDA,GABA,Gly) and neuropeptide(AT,AVP)increased,mRNA expressions of BDNF increased remarkably(P0.01) in Ganmai Dazao Decoction group and Venlafaxine group.Conclusion:Supplemented Ganmai Dazao Decoction and Venlafaxine increase mRNA expressions of receptor subtypes of monoamine,amino acid and neuropeptide,improve functions of cell signal transduction,increase expressions of BDNF in hippocampus to correct behavioral changes of rat model with depression.
