摘 要:采用响应面分析方法优化毛霉菌B的发酵培养基,首先通过单因素试验筛选出葡萄糖为最适碳源,酵母膏和玉米浆为最适氮源,用Plackett-Burman试验对葡萄糖、酵母膏、玉米浆、MgSO_4、FeSO_4、NH_4 Cl、K_2 HPO_4进行评估并筛选出具有显著效应的3个因素:葡萄糖、酵母膏、玉米浆,再通过最陡爬坡试验逼近其最大响应区域,最后采用B0x-Behnken试验对其用量进行优化,得到毛霉菌最佳发酵培养基(g/L):葡萄糖51.54,酵母膏5.22,玉米浆14.31,MgSO_4 0.5,FeSO_40.1,NH_4Cl 3,K_2HPO_4 3,pH 6.0~6.5。培养基优化后,毛霉生物量由23.51g/L提高至31.13 g/L,比对照组提高32.41%,腺嘌呤转化率由53.59%提高至59.97%,ATP产率由6.56 g/L提高至7.34 g/L,比对照组提高11.89%。Response surface methodology (RSM) was used to optimize fermentation medium for ATP production by Mucor. In single factor experiment, glucose was screened out as the most suitable carbon source, and yeast extract and corn steep liquor were screened out as the most suitable nitrogen sources. Plackett-Burman design assay was used to evaluate seven factors related to Mucor fermentation medium and three factors including glucose, yeast extract and corn steep liquor had statistically significant effects. Then steepest ascent procedures were applied to define optimal response re- gion of these three factors, finally the optimal factor concentrations were determined by adopting RSM analysis. The results showed that glucose 51.54 g/L, yeast extract 5.22 g/L, corn steep liquor 14.31 g/L, MnSO4 0.5 g/L, FeSO4 0.1 g/L, NH4 CI 3 g/L, K2 HPO43 g/L were combined as the most suitable medium for Mucor fermentation, which increased the productivity of Mucor from 23.51 g/L to 31.13 g/L, the conversion rate of adenine from 53.59% to 59.97 % and the productivity of ATP from 6.56 g/L to 7.34 g/L, respectively.
