非小细胞肺癌组织中核因子-κB-p65 mRNA及基质金属蛋白酶-1的表达及其与肿瘤转移的相关性

摘  要:目的探讨核因子-κB-p65(Nuclear factor-κB-p65,NF-κB-p65)与基质金属蛋白酶-1(Matrix metalloproteases-1,MMP-1)在非小细胞肺癌(Non-small cell lung cancer,NSCLC)组织中的表达及其与肿瘤浸润、转移、凋亡的相关性。方法收集50例手术切除的NSCLC组织及15例癌旁正常组织标本,RT-PCR法检测NSCLC及癌旁正常组织中NF-κB-p65 mRNA的表达,免疫组化法检测MMP-1蛋白的表达,TUNEL法检测细胞凋亡指数。结果 NSCLC组织中NF-κB-p65 mRNA的表达量显著高于癌旁正常组织(P<0.05);在腺癌组织中的表达量显著高于鳞癌组织(P<0.05);NSCLC组织中MMP-1蛋白阳性率显著高于癌旁正常组织(P<0.01);NF-κB-p65 mRNA和MMP-1蛋白的表达与TNM分期、分化程度、淋巴结转移、病理类型有关(P<0.05或<0.01),与年龄、性别无关(P>0.05);直线相关分析发现,NSCLC组织中NF-κB-p65 mRNA与MMP-1蛋白的表达相关;NSCLC组织中的细胞凋亡指数显著低于癌旁正常组织,且与NF-κB-p65 mRNA的表达量呈负相关(r=-0.547,P<0.001)。结论 NSCLC组织中NF-κB-p65 mRNA与MMP-1蛋白的表达具有相关性,NF-κB-p65 mRNA的表达与细胞凋亡指数呈负相关,NF-κB-p65通过一定的途径调控MMP-1蛋白的表达,NF-κB与MMP-1共同参与了肿瘤发生、发展的过程,NF-κB-p65发挥抗凋亡作用。Objective To investigate the expressions of nuclear factor (NF)-KB-p65 mRNA and matrix metalloprotease-1 (MMP-1) in non-small cell lung cancer (NSCLC) tissue and their correlation to the infiltration, metastasis and apoptosis of tumor. Methods Fifty specimens of NSCLC tissue and 15 specimens of normal lung tissue adjacent to tumor were collected from surgery, then determined for expressions of NF-KB-p65 mRNA and MMP-1 by RT-PCR and immunohistochemical assay respectively, and for apoptosis index by TUNEL method. Results The expression level of NF-KB-p65 mRNA was significantly higher in NSCLC tissue specimens than in normal lung tissue adjacent to tumor (P 〈 0. 05), while in adencarcinoma tissue than in squamous carcinoma tissue (P 〈 O. 05). However, the positive rate of MMP-1 in NSCLC tissue was significantly higher than that in normal lung tissue adjacent to tumor (P 〈 O. 01). The expressions of NF-KB-p65 mRNA and MMP-1 were correlated to the TNM stage, differentiation, metastasis in lymph node and pathological type of NSCLC (P 〈 O. 05 or P 〈 0. 01 ), while showed no correlation to the age and sex of patients (P 〉 0. 05). Linear correlation analysis showed correlation between the expressions of NF-KB-p65 mRNA and MMP-1 in NSCLC tissue. The cell apoptosis index was significantly lower in NSCLC tissue than in normal lung tissue adjacent to tumor (P 〈 O. 05), and was negatively correlated to the expression level of NF-KB-p65 mRNA (r = -0. 547,P 〈 O. 001). Conclusion In NSCLC tissue, the expression of NF-KB-p65 mRNA was correlated to that of MMP-1, and was negatively correlated to cell apoptosis index. NF-KB-p65 regulated the expression of MMP-1 protein by a certain route, and both NF-KB and MMP-1 involved in the onset and progress of tumor, in which NF-KB-p65 played an antiapoptotic role.
