健脾解毒中药胃肠安对人胃癌裸鼠皮下移植瘤生长及ERK1/2表达作用

摘　　要：目的:观察健脾解毒胃肠安对人胃癌SGC-7901裸鼠移植瘤生长、转移及p-ERK1/2蛋白表达影响。方法:人胃癌SGC-7901细胞BALB/C裸小鼠皮下移植瘤模型,随机分为3组,每组10只。对照组予生理盐水0.3mL灌胃,1次/d;胃肠安组予胃肠安煎剂0.3mL灌胃,1次/d;5-Fu组予5-Fu 2mg腹腔注射,1次/周。观察各组裸小鼠移植瘤生长与转移情况,免疫组织化学法检测各组移植瘤组织p-ERK1/2蛋白表达情况。结果:胃肠安组、5-Fu组与对照组相比,裸小鼠移植瘤体积、质量,肺转移结节数差异均有统计学意义(P<0.05);胃肠安组与5-Fu组及对照组相比,裸小鼠移植瘤组织p-ERK1/2蛋白表达较低,差异有统计学意义(P<0.01)。结论:胃肠安可以抑制SGC-7901胃癌生长、转移,可能与抑制p-ERK1/2相关。Objective ： To investigate the effects of Weichangan Decoction, a Chinese Herbal Formula, on tumor growth and metastasis and p- ERK1/2 expression in gastric carcinoma. Methods ：Thirty BALB/C nude mice transplanted with SGC- 7901 cells were randomly divided into three groups （10 mice per group） , and intragastric administered with normal saline or Weichangan Decoction daily,or i. p. administered with 5 - FU weekly. Tumor volume and metastatic nodules in lung were observed. Expressions of p - ERK1/2 proteins in the tumor tissues were detected by immunohistochemistry. Results ： Weichangan Decoction and 5 - Fu significantly reduced tumor volume, quality and number of metastatic nodules compared with controls（ P 〈 0.05）. The expressions of p - ERKI/2 proteins in tumor tissues in the Weichangan Decoction group were lower than those in the 5 - Fu and control groups （P 〈 0.01 ）. Conclusion：Weichangan Decoction has inhibitory effects on tumor growth and metastasis in SGC -7901 gastric carcinoma, and may be related to downregulation of p -ERK1/2 expression.
