摘  要:目的对催乳素(PRL)瘤进行体外细胞培养并进行激素分泌功能测定,从而为研究PRL瘤的病因、发病机制,为诊断和治疗PRL瘤提供实验基础。方法选取手术切除的垂体PRL腺瘤组织,采用悬浮细胞培养法,定时观察垂体腺瘤细胞的生长情况,并进行激素分泌功能测定。结果培养的PRL瘤细胞的特征:倒置相差显微镜下观察,原代培养的PRL腺瘤细胞呈椭圆形或梭形,24h后有部分PRL瘤开始半贴壁生长,大部分呈悬浮生长。于培养的约第10天开始,成纤维细胞数量开始增多,并逐渐取代PRL瘤细胞。PRL瘤细胞激素分泌功能测定:各例PRL腺瘤培养细胞PRL分泌量第1~5天逐渐增加,第6~8天达到高峰。测定结果显示培养的PRL瘤细胞在第7天功能状态最佳。结论通过本次试验研究,可以为以后治疗PRL腺瘤提供良好的试验基础,可以通过干预垂体腺瘤细胞,观察PRL腺瘤细胞的生长及激素分泌特征,为研究PRL腺瘤的发病机制、药物治疗及预后具有重要意义。Objective Of prolactin (PRL) tumors in vitro cell culture and for determination of hormone secretion, and thus for the study of PRL tumor etiology, pathogenesis, diagnosis and treatment for the PRL tumors provide an experimental basis. Method Select the pituitary PRL adenomas resected tissue, using the suspension cell culture, regularly observe the growth of pituitary adenoma cells, and the determination of hormone secretion. Results PRL cells cultured tumor characteristics: inverted phase contrast microscope, primary cultures of PRL adenoma cells were oval or spindleshaped, 24h after the start of some semi-adherent PRL tumor growth, most of the growth in suspension. Pituitary adenoma was the growth of groups, or flake. In the culture around day 10, began to increase the number of fibroblasts and tumor cells gradually replace the PRL. PRL secretion of tumor cell hormone function test: the cases of PRL secretion ofPRL adenoma cell cultures 1 to 5 days a gradual increase in the first 6 to 8 days to reach the peak. Measured results show that PRL tumor cells cultured 7 days in the best functional status. Conclusions Through this pilot study for the future treatment of PRL adenomas can provide a good experimental basis, can interfere with pituitary adenoma cells were PRL adenoma cell growth and hormone secretion characteristics for the study of PRL adenoma pathogenesis, drug treatment and prognostic significance.
