摘 要:目的:建立高效液相色谱(high performance liquid chromatography,HPLC)法测定牛奶中胆固醇的方法。方法:色谱柱为Shimadzu C18柱(4.6mm×250mm,5μm),流动相为乙腈,流速为1.0mL/min,检测波长为203nm,柱温为30℃。结果:胆固醇的线性范围为3.58~358μg/mL(r=0.9999),平均回收率为101.8%(n=6),相对标准偏差为1.14%。结论:该方法测定结果准确、重现性好,可作为测定牛奶中胆固醇含量的方法。Objective: To establish an HPLC method for the determination of cholesterol in milk.Methods: Cholesterol was separated on a Shimadzu C18column(4.6 mm × 250 mm,5μm) using acetonitrile as mobile phase at a flow rate of 1.0 mL/min.The detection wavelength was set at 203 nm and the column temperature at 30 ℃.Results: A calibration curve was established revealing a good linear relationship in the range of 3.58-358μg/mL with a correlation coefficient of 0.9999.The average spike recovery rate was 101.8% with a RSD of 1.14%.Conclusion: The method was accurate,reproducible and applicable for the determination of cholesterol in milk.
