摘 要:目的:观察红光照射对急性闭合性软组织损伤大鼠损伤组织中细胞因子白介素-1β(interlenkin-1β,IL-1β)、前列腺素E2(postaglandin E2,PGE2)的浓度和机械痛敏的影响,评价其对肌肉损伤后炎症和疼痛治疗的作用。方法:健康的雄性Wistar大鼠66只,随机分为正常对照组(n=6)、自然愈合组(n=30)和红光照射组(n=30)。采用自制的"重物自由落体"打击装置通过单次撞击建立大鼠急性腓肠肌损伤模型。造模前和造模后第1,2,3,5和7天分别用von Frey细丝测量大鼠机械缩足反射阈值(mechanical withdrawal threshold,MWT)的变化;并于造模后第1,2,3,5和7天通过ELISA测定受损组织中细胞因子IL-1β和PGE2的含量。结果:自然愈合组大鼠损伤后各时间点MWT与红光照射组MWT相比均下降更明显(P<0.01);损伤后第1,2,3,5和7天,自然愈合组大鼠损伤组织中细胞因子IL-1β和PGE2的表达量各时间点均显著高于正常对照组和红光照射组大鼠(P<0.01,P<0.05)。结论:红光照射治疗明显降低了损伤组织中促炎和致痛细胞因子IL-1β、PGE2的表达,从而证明其具有抗炎、镇痛作用。Objective:To investigate the impact of red light therapy(RLT) on concentrations of cytokines interleukin-1 beta(IL-1β) and prostaglandin E2(PGE2) in acute closed-soft-tissue injured rats,and to evaluate the effects of RLT on inflammatory response and pain after injury.Methods: 66 male Wistar rats were randomly divided into three groups: control group(uninjured,n = 6),injured-non-RLT group(n = 30),injured-RLT group(n = 30).Blunt trauma to the gastrocnemius muscle of rat was produced by single strike delivered by custom-designed "free-falling-mass" device.Mechanical withdrawal threshold(MWT) was measured respectively using the von Frey filaments on 1,2,3,5 and 7 days after injury.Furthermore,at the corresponding time points,the concentration of cytokines IL-1β and PGE2 in injured tissue were detected by enzyme-linked immunosorbent assay(ELISA).Results: The MWT of rats in injured-nonRLT group reduced notably after injury compared with that of the injured-RLT group at all time points(P 0.01).The expression of IL-1β and PGE2 in the injured tissue of rats in the injured-non-RLT group was significantly higher than that in the control and injured-RLT groups(P 0.01 and P 0.05,respectively).Conclusion: RLT can significantly reduce the expression of proinflammatory and pronociceptive cytokines IL-1β and PGE2,suggesting that RLT has effects of anti-inflammation and analgesia.
