白细胞介素-1β基因多态性与人胃癌发病风险及病理特征的关系研究

摘　　要：目的研究白细胞介素（IL）-1β基因多态性与人胃癌发病风险及病理特征的关系。方法选择2010年1—12月经病理确诊为胃癌标本200例（病例组），慢性浅表性胃炎标本200例（对照组），提取DNA后应用基因芯片技术检测IL-1β基因-511、-31、-1473、＋3954位点的多态性。观察IL广1β基因-511、-31、-1473、＋3954位点的多态性与胃癌发病风险及临床病理参数的关系。结果病例组IL-1β基因-511、-31、-1473、＋3954位点的T等位基因频率分布分别为48．75％（195／400）、55．25％（221／400）、53．25％（213／400）、50．75％（203／400），与对照组的47．25％（189／400）、53．00％（212/400）、52．50％（210／400）、52．50％（210／400）比较差异有统计学意义（P〈0．05）。IL-1β基因-511、-31位点的T等位基因胃癌分化程度较低，IL-1β基因-511、-1473位点的T等位基因胃癌发生年龄相对较早。结论IL-1β基因-511、-31、-1473、＋3954位点基因型增加胃癌发病的风险。IL-1β基因-511、-31位点的T等位基因与胃癌的分化程度有关，IL-1β基因-511、-1473位点的T等位基因与胃癌患者的年龄具有相关性。Objective To investigate the correlation between polymorphism of interleukin （IL）-1β genes and risk and pathological characteristics of gastric cancer in human. Methods From January to December 2010, 200 cases of gastric cancer （patient group） and 200 cases of chronic superficial gastritis （control group） were collected. DNA was extracted and IL-1β gene -511,-31 ,-1473,＋3954 site were detected by gene chip technology. The correlation between IL-1β gene -511,-31,-1473, ＋3954 site and risk and pathological characteristic of gastric cancer was observed. Results The genotype frequency of IL-1 13 gene-511 ,-31 ,-1473,＋3954 site was 48.75%（195/400） ,55.25%（221/400）,53.25%（213/400）, 50.75% （203/400） in patient group, 47.25% （189/400）,53.00% （212/400）,52.50% （210/400）,52.50% （210/400） in control group, and there was significant difference between two groups （P 〈 0.05）. While IL-1β gene -511,-31 site T allelic with the lower degree of differentiation of gastric cancer, IL-1β gene -511 ,-1473 site T allelic with the early age of gastric cancer. Conclusions IL-1β gene -511 ,-31, -1473,＋3954 site genotype increase the risk of gastric cancer. IL-1β gene -511 ,-31 site T allelic are related with the degree of differentiation of gastric cancer. IL-1β gene -511,-1473 site T allelic are related with age of gastric cancer patient.
