摘 要:目的:探讨微弧氧化(micro-arc oxidation,MAO)改性后的纯钛对成骨细胞(MG63)蛋白合成功能的影响。方法:本实验分为实验组和对照组,实验组对钛片进行微弧氧化(MAO)处理,对照组仅对钛片进行抛光处理。采用电镜观察试样表面形貌,将MG63细胞与钛材复合培养,通过MTT方法检测成骨细胞增殖,并测定碱性磷酸酶(Alkaline phosphatase,ALP)活性。通过蛋白质印迹法(Western-blot)检测各组成骨细胞的成骨相关蛋白:I型胶原(Collagen-I,Col-I)、骨钙素(osteocalcin,OCN)、骨桥蛋白(osteopontin,OPN)、runt相关转录因子2(runt-related transcription factor-2,Runx-2)的表达水平。结果:经MAO处理后,纯钛表面形成一层多孔结构。MG63细胞在MAO处理钛表面3、7d的增殖活性及ALP活性高于对照组(P<0.05)。3d时,实验组Col-1、OCN、OPN、Runx-2的蛋白表达水平明显高于对照组(P<0.05)。结论:纯钛经MAO处理后,在表面形成多孔复合的形态特征,能促进成骨细胞的增殖及分化,并显著提高成骨细胞成骨相关蛋白的表达水平。Objective: To evaluate the effect of titanium modified by micro-arc oxidation (micro-arc oxidation, MAO) on initial response of osteoblasts, especially the protein synthesis. Methods: Two groups were involved in this study, MAO and control group (smooth titanium). Surface morphologies were observed by scanning electron microscope. Proliferation and alkaline phosphatase activity were carried out in vitro tests. In addition, protein syntheses, including Col-Ⅰ, OCN, OPN and Runx-2, was evaluated by western blotting test. Results: A porous layer was observed on titanium surface modified by MAO. Initial osteoblast proliferation and ALP activity were enhanced significantly on the micro-arc oxidized surfaces compared with that on smooth surface (P〈0.05). After cultured for 3d, Col-1, OCN, OPN, Runx-2 protein expression were increased (P〈0.05) in MAO group compared with that in control group. Conclusion: Titanium surface modified by MAO could promote the proliferation, differentiation and protein synthesis of osteoblasts.
