波动性高糖下血红素加氧酶1对INS-1细胞氧化应激的影响

摘  要:目的探讨波动性高糖下血红素加氧酶1(HO-1)对INS-1细胞氧化应激的影响。方法以体外培养的INS-1细胞为研究对象,分为正常对照组、持续性高糖组、波动性高糖组、钴原卟啉(CoPP)+波动性高糖组、锌原卟啉(ZnPP)+波动性高糖组。各组均干预72h,Western blot检测HO-1蛋白表达水平,流式细胞仪测定活性氧簇(ROS)水平,四氮唑蓝(MTT)法测定细胞活力。结果波动性高糖组HO-1蛋白表达及ROS水平均显著高于持续性高糖组(P均<0.01),波动性高糖组细胞活力显著低于持续性高糖组(P<0.05);与波动性高糖组相比,CoPP+波动性高糖组HO-1表达水平及细胞活力升高、ROS水平下降(P均<0.05),而ZnPP+波动性高糖组呈现相反的变化趋势。结论波动性高糖较持续性高糖引起INS-1细胞产生更严重的氧化应激,在波动性高糖条件下,HO-1可能通过抗氧化应激对INS-1细胞发挥保护效应。Objective To study the effects of heme oxygenase-1(HO-1) on the oxidative stress of INS-1 cells under fluctuant high glucose. Methods INS-1 cells were cultured and divided into five groups: control,persistent high glucose(PHG),fluctuant high glucose(FHG),CoPP + FHG,and ZnPP+ FHG.After 72 hours of intervention,the expressions of HO-1 protein were determined by Western blot,the intracellular reactive oxygen species(ROS) were detected by flow cytometry,and the cell viabilities were evaluated by MTT assay. Results The HO-1 expressions and ROS levels were higher in the FHG group than in the PHG group(P〈0.01,respectively).The cell viabilities were significantly lower in the FHG group than in the PHG group (P〈0.05).In comparison with FHG,CoPP+ FHG had greater HO-1 protein expressions and cell viabilities,but lower ROS levels(P〈0.05,respectively),and the ZnPP+FHG had lower HO-1 protein expressions and cell viabilities,but greater ROS levels(P〈0.01,respectively). Conclusions Fluctuant high glucose may trigger more severe oxidative stress in INS-1 cells than persistent high glucose and HO-1 may protect INS-1 cells against oxidative stress under fluctuant high glucose.
