摘 要:目的:建立虎杖指纹图谱与抗氧化活性之间的谱效关系.方法:采用Zorbax SBC柱(250mm×4.6mm,5μm)色谱柱,流速1.0mL/min;波长280nm;流动相A为水,流动相B为甲醇,梯度洗脱程序为0~5min,B泵15%~30%;5~45min,B泵30%~70%;45~50min,B泵70%~80%;50~70min,B泵80%~100%.采用清除1,1-二苯基苦基苯肼(DPPH)自由基法对不同质量浓度的虎杖提取物进行抗氧化活性测定.结果:13批药材的指纹图谱被得到,其中12个特征共有峰被确定,采用中国药典指纹图谱相似度评价软件进行了相似度计算,并测定了其抗氧化活性,虎杖提取物在质量浓度为0.23mg/mL时,其自由基清除率最高93.7%.初步确定与其药效具有显著性的特征指纹峰为峰3、10、11和12.结论:本研究中建立的虎杖指纹图谱中与抗氧化活性相关的主要活性成分,为虎杖药材的药用物质基础研究、提高其质量标准提供了可靠依据,也为其他中药材质量标准的提高奠定了基础.Objective:To investigate the relationship between fingerprints of Polygonum cospidatum and the antioxidant activities of its water extract.Methods:HPLC(high-performance liquid chromatography) was used to establish the fingerprints of P.cospidatum.A Zorbax SBC(250 mm×4.6 mm,5 μm) column was used.The flow rate was 1.0 mL/min.The mobile phase consisted of water(A) and methanol(B),using a gradient program of 15%-30%(B) in 0-5 min,30%-70%(B) in 5-45 min,70%-80%(B) in 45-50 min and 80%-100%(B) in 50-70 min.The wavelength was set at 280 nm.The antioxidant activities were determined by DPPH radical scavenging rate.Results:The fingerprints of 13 batches of the medicine were obtained and 12 characteristic common peaks were detected,and the correlation coefficient of similarity in chromatograms based on the peak areas of characteristic compounds were calculated for quantitative expression of the HPLC fingerprints by the professional similarity evaluation software(Chinese Pharmacopoeia Commission).It was found that the extract of P.cospidatum had good scavenging effects on free radicals.With a mass concentration of 0.23 mg/mL,the extract showed a maximum scavenging rate of 93.7%.It was tentatively concluded that Peaks 3,10,11 and 13 were its characteristic peaks.Conclusion:Close correlation existed between the spectrum-effect relationships based on the HPLC fingerprints and antioxidant activity.The results also provide a basis for further quality control of P.cospidatum.
