摘  要:目的筛选汉坦病毒β3整合素受体拮抗肽,为进一步设计和获取高活性的抗病毒短肽提供参考资料。方法以郎整合素为靶分子,从随机七肽噬菌体展示库中筛选β3整合素的高亲和力噬菌体配体,应用相关软件对阳性噬菌体的基因DNA测序结果进行分析。结果通过比较亲和性噬菌体外源插入肽的DNA序列,发现LPLTPLP、TGVKGPG、DHRNHLV肽段有较高的重复概率,经与汉滩病毒囊膜糖蛋白氨基酸序列比较发现,两者存在一定的同源序列。结论运用噬菌体展示技术从肽库中筛选邸整合素受体拮抗剂是可行的。Objective To screen and identify the antagonists against β3 integrin, the receptor of Hantavirus, from a heptapeptide phage display library, so as to provide new data for designing high activity antivirus peptides. Methods The integrin-binding phage clones were screened from 7-met phage display library using β3 integrin as target protein. The sequences of single strand DNA sequences were determined and analyzed. Results According to DNA sequence, three peptides, LPLTPLP, TGVKGPG and DHRNHLV were found. The three peptides had high repetition rate. The comparison of the amino acid sequences of foreign peptides of the clones to HTNV glycoprotein shows some similarity. Conclusion The antagonists of β3 integrin receptor could be selected from phage display library.
