改良的血管端端吻合法建立大鼠原位肾移植模型

摘　　要：目的采用改良的血管端端吻合法建立一种简单、稳定、可靠的大鼠原位肾移植模型。方法利用wistar大鼠作为供体和受体,取供鼠的双肾作为供肾,原位低温灌注,显微外科技术行改良的二定点顺序缝合法端端吻合肾动脉,后壁-前壁缝合法连续缝合肾静脉,进行左侧原位异体肾移植,供体输尿管与受体输尿管端端吻合重建尿路。结果完成大鼠肾移植模型40次,成功率为82.5%。肾动脉端端吻合时间(11±2min),肾静脉吻合时间(7±2min),输尿管端端吻合时间(12±2min),供体手术时间(30-40min),供肾缺血时间(40-60min),手术时间共约(120±20min)。结论采用改良的血管端端吻合法重建血管具有缝合确切,易于掌握、简便、可靠等优点,通过技术改良成功建立了一种可靠、成功率高、结果稳定的大鼠原位肾移植模型。Objective To create a simple, stable and reliable model of orthotopic kidney transplantation by using modified vascular anastomosis technique. Methods Wistar rats as donor and recipient,Two renal allografts were havested from one donor, with low temperature saline perfusion. Then transplantation of kidney in situ were performed,in which microsurgical anastomosis of renal arte,posterior- the former end to end anastomosis renal vein,and end to end anastomosis ureter in order to rebuilt urinary passage. Results Forty kidney transplant models were completed, with success rate 82.5~. The time for renal artery anastomosis was 11-1-2 rain,the renal vein anastomosis time 7-t-2 min,u- reter anastomosis time 12±2 min, the donor operation time 30-40 min, for renal donor renal ischemia time 40- 60 min,operation time of about 120±20 min. Conclusion The Modified method of vascular anastomosis with reconstruction of vascular anastomosis was accurate,easy tO grasp, simple and reliable thai a modified model of orthotopic kidney transplantation was easily accessible by using this improved technique.
