摘 要:目的探讨雌激素受体(ER)表达不同的乳腺癌细胞间相互作用。方法 PCR法检测MDA-MB-231及MCF-7细胞中ER的基因表达。CCK-8法检测17β-雌二醇对MCF-7和MDA-MB-231细胞生长增殖的影响。将MDA-MB-231(雌激素受体阴性细胞)用羧基荧光素二醋酸盐琥珀酰亚胺酯(CFSE)法标记荧光信号,将其分别与MCF-7细胞(雌激素受体阳性细胞)和未标记荧光的MDA-MB-231细胞(雌激素受体阴性细胞)用Transwell侵袭小室分层共培养,加入或不加入雌二醇,检测MDA-MB-231细胞的增殖差异。结果 MCF-7细胞表达雌激素受体α(ESR1,ERα)和雌激素受体β(ESR2,ERβ),而MDA-MB-231细胞均不表达。17β-雌二醇浓度为10-11 mol/L时促MCF-7增殖作用最大,与对照组相比差异具有统计学意义(P<0.05),对MDA-MB-231细胞则无明显作用(P>0.05)。在加入雌二醇培养的MCF-7细胞与MDA-MB-231细胞小室分层共培养组,经流式细胞仪检测发现MDA-MB-231平均荧光强度较对照组显著降低。结论雌激素受体阳性细胞在雌激素作用下,可促进雌激素受体阴性细胞的增殖。Objective To detect the interaction between ER positive breast cancer cells and ER negative breast cancer cells.Methods The expression of ER in MDA-MB-231 and MCF-7 cells was detected by using PCR.The effects of estrogen on growth and proliferation of MCF-7 and MDA-MB-231 cells were tested by CCK-8 method.MDA-MB-231 cells were labeled with CFSE,and co-cultured with MCF-7 cells in Transwell chamber.The proliferation of ER negative cells labeled with CFSE was observed in the presence or absence of estrogen by using flow cytometry.Results MCF-7 cells expressed ESR1 and ESR2,while MDA-MB-231 cells expressed none.10-11 mol/L 17β-oestradiol could promote the proliferation of MCF-7 cells(P0.05),but had no obvious influence on MDA-MB-231 cells(P0.05).The mean fluorescence intensity of MDA-MB-231 cells labeled with CFSE was decreased significantly when co-cultured with MCF-7 cells and estrogen.Conclusion ER positive cells cultured with estrogen could stimulate the proliferation of ER negative cells.
