胃乐散对大鼠胃溃疡黏膜EGF、EGFR和TGFβ_1的影响及意义

摘　　要：目的观察胃乐散对大鼠实验性胃溃疡表皮生长因子(EGF)、表皮生长因子受体(EGFR)及转化生长因子β1(TGFβ1)的影响,并探讨其作用机理。方法 SD大鼠48只,随机分为正常对照组、模型对照组、胃乐散低剂量组、胃乐散中剂量组、胃乐散高剂量组和雷尼替丁组,采用冰醋酸烧灼法制备大鼠胃溃疡模型,连续用药后第14天处死大鼠,取溃疡部位及周围组织提取蛋白,用免疫印迹法检测组织中EGF、EGFR、TGFβ1的含量。结果与正常对照组比较,模型组EG-FR、TGFβ1水平降低(P<0.05),胃乐散高剂量组及雷尼替丁组EGFR升高(P<0.01),胃乐散中、高剂量组的TGFβ1含量高于正常对照组(P<0.01),而模型组EGF的含量较正常对照组增加(P<0.05),但胃乐散各组及雷尼替丁组增加更为明显(P<0.01)。与模型组比较,胃乐散中、高剂量组及雷尼替丁组EGFR、TGFβ1及EGF的表达均显著提高(P<0.01)。结论胃乐散治疗胃溃疡可能是通过促进细胞因子TGFβ1的分泌及促进胃黏膜中EGF和EGFR的表达实现的。Objective To investigate the effects of Wei-Le-San on epidermal growth factor（EGF）,epidermal growth factor receptor（EGFR） and transforming growth factor β1（TGFβ1） in gastric ulcers of rats,and probe its functional mechanism. Methods 48 rats were randomly divided into normal group,model group,low dose of Wei-Le-San group,middle dose of Wei-Le-San group,high dose of Wei-Le-San group and ranitidine group.The rats of the gastric ulcer model were used by the glacial acetic acid.After drug treatment for 14 days,the rats gastric mucosa were collected to check the content of EGF、EGFR and TGFβ1 with immunoblot（western blotting）. Results Compared to the control,the levels of EGFR and TGFβ1 in model group,significantly derceased（P0.05）,and the content of EGF increased（P0.05）.But the increase of Wei-Le-San groups and ranitidine group was more obvious（P0.01）.Compared to the model group,Wei-Le-San elevated the level of EGF、 EGFR and TGFβ1 in Gastric ulcer（P0.01）. Conclusion One of the possible mechanism is Wei-Le-San promotes secretion of the cytokines TGFβ1 and increases expression of EGF and EGFR to cure the gastric ulcer.
