摘 要:目的 为比较不同肿瘤细胞株诱导分化时表面N-糖链结构的变化。方法 采用 Con A和 DSA凝集素亲和柱层析法分析 SMMC 772 1人肝癌细胞受 ATRA和 db-c AMP以及 HL - 6 0人早幼粒白血病细胞受 ATRA和 PMA诱导分化时细胞表面糖蛋白上 N-糖链的结构变化。结果 ATRA和 db- c AMP均使 SMMC 772 1细胞表面高甘露糖型/杂合型和三 /四天线复杂型 N-糖链减少而二天线复杂型N-糖链增多。ATRA和 PMA也均使 HL- 6 0细胞表面三 /四天线 N-糖链减少和二天线 N-糖链增加 ,但却使高甘露糖型/杂合型 N-糖链增加。结论 同一细胞受不同诱导剂分化时 ,其表面 N-糖链的结构有相同的变化 ,也有不同的变化 ;Objective To compare the changes in the structure of N glycans on the cell surface during the induced differentiation of different tumor cell lines. Methods The structural changes of surface N glycans on human hepatocellular carcinoma cell line, SMMC 7721, induced by all trans retinoic acid (ATRA) or dibutyryl 3', 5' cyclic adenosine monophosphate (db cAMP), as well as on acute promyelocytic leukemia cell, HL 60, induced by ATRA or phorbol 12 myristate 13 acetate (PMA) were analyzed by using ConA and DSA lectin affinity column chromatographies.Results It was found that the content of high mannose type/hybrid type N glycans and tri /tetra antennary complex type N glycans were decreased, while biantennary complex type N glycan was increased on SMMC 7721 cell surface by ATRA or db cAMP treatment. The content of tri /tetra antennary complex type N glycans was decreased and the biantennary complex type N glycan was increased on the surface of HL 60 cells by the treatment of ATRA or PMA, but the content of high mannose type/hybrid type N glycan was increased by these treatments, the change was just opposite to that of SMMC 7721 cells. Conclusion These results suggest that the structure of surface N glycans shows not only the same changes, but also some different changes in the induced differentiation of same cell line by different compounds or of different cell line by same compound.
