去甲基斑蝥素治疗卵巢癌的实验研究

摘　　要：目的　观察去甲基斑蝥素 (NCTD)体内外对人卵巢癌细胞株 3AO和 AO的抑制作用。方法　利用细胞培养技术 ,以结晶紫染色法测定 NCTD对 3AO和 AO细胞的体外杀伤作用 ,并在光镜和电镜下观察 NCTD对其进行治疗 ,以观察 NCTD体内对卵巢癌的抑癌效应。结果　体外实验表明 NCTD对 3AO和 AO细胞有较强抑制作用 ,并呈浓度和时间依赖性。在 NCTD作用下 ,光镜下卵巢癌细胞大多变圆呈悬浮状 ,电镜下则表现为细胞微绒毛消失 ,线粒体水肿、嵴断裂 ,胞质呈空泡样变性。体内实验则表明无论是腹腔内注射还是瘤内注射 ,NCTD对卵巢癌均有抑制瘤效应。结论　 NCTD体内外对卵巢癌均有较强的抑制作用 ,提示Objective To evaluate the inhibitory effects of Norcantharidine(NCTD) on human ovarian cancer 3AO and AO cell lines in vitro and in vivo.Methods Inhibitory effects of NCTD on 3AO and AO cells in vitro were measured by crystal violet dyeing;morphological and ultrastructural changes were observed by light and electron microscopes. An in vivo antitumor activity of NCTD,either ip or intratumoral administration,was studied in SCID mouse model with 3AO cells subcutaneously transplanted. Results NCTD had killing activity on 3AO and AO cells in vitro,and was dose and time dependent. Under light microscope,most of 3AO and AO cells turned round and suspended in medium. Under electronic microscope, cells changed with disappearing of microvillus, swelling and crestas breaking of mitochondria. NCTD also had antineoplastic activity on ovarian cancer transplanted to SCID mice.Conclusions In vivo and in vitro,NCTD showed strong inhibition on ovarian cancer cells and may be effective in clinical treatment of ovarian cancer cells and may be effective in clinical treatment of ovarian cancer.
