白及组培快繁的实验研究

摘　　要：目的:探索白及组培快繁的最适培养基和培养条件,为进一步完整建立白及快繁体系奠定基础。方法:以白及成熟蒴果为材料,在不同培养基上进行无菌播种,种子萌发后利用组织培养方法进行无性系繁殖。结果:白及种子萌发的最佳培养基为1/2 MS+6-BA 1.0 mg/L;白及丛生芽增殖最佳培养基MS+1.0mg/L BA+0.1mg/L NAA,1/2MS+1.0 mg/L NAA+7.5%香蕉汁有利于白及生根,MS+0.1mg/L NAA+1.0mg/L 6-BA+7.5%香蕉汁既可以促进白及球茎的生长,在一定程度上又可以促进白及的生根。结论:激素于白及组培快繁有显著影响,通过激素适当配比得到白及快繁的最佳培养体系,对白及资源的利用和保护有重要意义。Objective： To explore the best medium and tissue culture condition and pave the way for further establishing the rapid propagation system of Bletilla striata. Methods ： Capsular fruit of Bletilla striata was sowed in different medium in the sterile environment, then clonal reproduction of Bletilla striata used Tissue culture method. Results ： The best medium of seeds germination is 1/2 MS ＋ 6 - BA 1.6 mg/L. The best medium of proliferateing is MS ＋ 1. Omg / L 6 - BA ＋0. lmg / L NAA , 1/2MS ＋ 1.0 mg/L NAA ＋7.5% banana juice is benefit for the root growth of Blerilla striata. The medium of MS ＋0. lmg/L NAA ＋ 1.0mg/L 6 - BA ＋7.5% banana juice can promote the corm growth and roo ting growth. Conclusion： Hormone plays an important role in rapid propagation of Bletilla strata. It is found the best culture of Bletilla strata propagation system by appropriate ratio of cytokinin and growth regulators.
