摘  要:目的:探讨外周型苯二氮卓受体(PBR)在大鼠肝星状细胞(HSC)上的表达。方法:采用肝脏离体胶原酶灌注消化及密度梯度离心的方法来分离培养大鼠HSC;应用实时监测荧光定量PCR技术(RT-PCR),于造模第4、6、8周时检测大鼠HSC PBR mRNA的水平。结果:在第4、6、8周时模型组大鼠较同期正常组大鼠的HSC PBRmRNA的表达量明显增加(P<0.01);模型组大鼠随着造模时间的延长,HSC PBR mRNA的表达量逐渐增加,差异有显著性意义(P<0.01或P<0.01);而不同时期正常组大鼠HSC PBR mRNA的表达差异无显著性意义。结论:肝纤维化过程中,HSC PBR mRNA的表达水平显著增加。Objective:To investigate the changes of peripheral benzodiazepine receptors(PBR)expression in hepatic stellate cells(HSCs).Methods:Liver exvivo perfusion of collagenase and density gradient centrifugation were used to isolate hepatic stellate cell.The changes of PBR mRNA in hepatic stellate cells were observed by the real-time FQ-PCR method.The results were judged with circle number of times(CT) when the level of amplification exponent achieved detecting liminal value.Results:Comparing the model group with the healthy control at the 4th,6th,8th week,the changes of CT were significant.PBR mRNA expression was higher in the model group than that in the healthy control.Conclusion:In the process of hepatic fibrosis,HSC PBR expression level is significantly increasing.
