姜黄素对PC3和H446细胞中Id1mRNA表达的影响

摘  要:目的观察姜黄素对雄激素非依赖性前列腺癌PC3和小细胞肺癌H446细胞Id1mRNA表达的影响。方法分别采用MTT法检测细胞生长活性;划痕实验检测细胞迁移能力;Hoechst染色检测细胞凋亡;实时荧光RT-PCR法检测PC3和H446中Id1mRNA的表达。结果姜黄素明显抑制PC3和H446细胞中Id1mRNA的表达,其相对定量随姜黄素浓度增大而减少;同时PC3和H446细胞的生长抑制率和凋亡率增加,细胞迁移距离减少。结论在PC3和H446细胞中,姜黄素可能通过抑制Id1基因表达发挥其抑制细胞生长、迁移和诱导细胞凋亡的作用。[Objective ] To determine if curcumin affects the expression of IdlmRNA in androgen-independent human prostate cancer PC3 ceils and small cell lung cancer H446 cells. [Methods] The celt viability of PC3 and H446 was evaluated by MMT assay. The ability of cell migration was determined by a scratch wouml assay. The cell apoptosis was assayed by Hoechst staining. The quantity of Idl mRNA was detected by real time quantitative reverse transcription-polymerase chain reaction (RT-PCR). [Results] The relative quantity of IdhnRNA was significantly reduced by cureumin in PC3 and H446 cells. The quantity of IdlmRNA was inhibited by curcumin in a dose dependent fashion. Meanwhile, the cell inhibition rate and cell apoptosis rate were obviously increased by curcumin. The cell migrating distance was shorter in curcumin treated cells. [ Conclusion] The inhibition of IdlmRNA by eurcumin in PC3 and H446 cells is a novel revelation that may be one important mechanism of decreased cell viability, migration ability and induced cell apoptosis.
