四磨汤对胃窦平滑肌的影响及钙通道在其中的作用

摘　　要：目的研究四磨汤对SD大鼠离体胃窦纵、环行平滑肌条收缩活动的影响及钙通道在其中的作用。方法正常SD大鼠离体胃窦纵、环行平滑肌条置于盛有Krebs液的组织浴槽中,记录其等长收缩活动,观察四磨汤累积量加入(1μL、5μL、25μL、50μL、100μL、150μL、200μL)对大鼠胃窦纵、环行平滑肌条收缩活动的影响,以及钙通道阻断剂硝苯地平(30 nmol/L)对四磨汤引起的胃窦平滑肌条收缩活动的影响。结果四磨汤剂量依赖性引起大鼠胃窦纵、环行平滑肌条收缩活动增加,硝苯地平可部分阻断四磨汤对胃窦纵、环行平滑肌条的兴奋作用;硝苯地平对于四磨汤诱导的纵行肌兴奋的抑制效应明显强于环行肌。结论四磨汤对大鼠胃窦平滑肌的收缩活动具有明显的兴奋作用,这一兴奋作用部分通过钙通道介导,纵行肌较环行肌更多的依赖于外钙内流。Objective To study the effect of Simo Decoction on the contractile activity of isolated antral longitudinal and circular strips in SD rats and its underlying mechanism.Methods The contractions of antral longitudinal and circular strips in SD rats were noted in the organ bath under baseline or addition of nifedipine（30 nmol/L） respectively,and consecutive concentrations of Simo Decoction（1 μL,5 μL,25 μL,50 μL,100 μL,150 μL,200 μL） were put into the bath to observe the strip responses.The results of strip responses to Simo Decoction were expressed as the variational percent of area（integral）/min before and after adding each medication in the bath.Results Simo Decoction dose-dependently increased the contractions of antral longitudinal and circular strips.After incubation with calcium channel inhibitor nifedipine,the increased contractile activity induced by Simo Decoction in longitudinal and circular strips could be partly inhibited;the inhibitory effect of Simo Decoction on longitudinal strips was stronger than that on circular strips.Conclusion Simo Decoction can dose-dependently promote the contractions of antral longitudinal and circular muscles,which partly decided by activation of Ca2＋ channel;the stimulatory effect of Simo Decoction on longitudinal strips is more dependent on activation of Ca2＋ channel than that on circular strips.
