摘  要:目的探讨氯丙咪嗪对人脑胶质瘤U251细胞体外增殖的影响。方法用甲基噻唑基四唑(MTT)比色法检测不同浓度的氯丙咪嗪对U251细胞增殖活性的影响。根据结果,选取二个不同浓度组检测细胞形态学的变化、台盼兰染色细胞死亡计数、细胞凋亡染色、流式细胞术检测等。结果依据MTT实验结果,选取氯丙咪嗪57μmol/L和28.5μmol/L两个浓度进行台盼兰染色、细胞形态学、细胞凋亡染色和流式细胞术检验。结果证实,氯丙咪嗪对U251细胞有明显的抑制作用。结论氯丙咪嗪具有明显的抑制胶质瘤U251细胞增殖的功能,促进其凋亡,可作为胶质瘤化疗的备选药物。Objective To investigate the effects of chlorimipramine on the growth of human brain U251 glioma cells in vitro. Methods Using methyl thiazolyl tetrazolium(MTT) colorimetry to test the effects of different concentrations of clomipramine on the growth of human brain U251 glioma cells were studied with MTT colorimetry in vitro, based on which two group with different concentrations of clomipramine were selected for being examined by cell morphology, death count of trypan blue staining cells, apoptosis staining, and flow cytometry. Results The results from the tests on clomipramine 57 μmol/L and 28. 5 bμmol/L showed that clomipramine significantly inhibited the growth of human brain U251 glioma cells in vitro. Conclusion Clomipramine can significantly inhibit the growth and promote the apoptosis of human brain U251 glioma cells in vitro and may be taken as a second choice of antiglioma drug for chemotherapy.
