摘  要:目的检测乙型肝炎病毒(HBV)感染者精子中去唾液酸糖蛋白受体(ASGP R)的表达,并与HBV阴性男性精子比较,探讨ASGP-R是否参与介导HBV侵入人精子。方法用ASGP-R单抗,通过免疫印迹(Western blot)和流式细胞术检测ASGP-R在乙肝表面抗原(+)/乙肝e抗原(+)[HBsAg(+)/HBeAg(+)]者(双阳性组,n=16)、HBsAg(+)/HBeAg(一)者(单阳性组,n=28)和HBsAg(一)者(阴性组,n=35)精子中表达差异。结果 ASGP-R于人精子的胞质中高表达,于胞膜低表达;胞质中ASGP-R水平在阴性组、单阳性组及双阳性组中逐渐升高,双阳性组和单阳性组的胞膜上ASGP-R的阳性率均高于阴性组(P均<0.05)。结论人精子胞膜和胞质中均存在ASGP-R;HBV可能通过人精子表面的ASGP-R进入精子,目HBV可能通过某种机制使ASGP-R的表达量增加。Objective: To explore the mechanism of hepatitis B virus (HBV) invading into human sperm through detecting and comparing the expression of the asialoglycoprotein receptor(ASGP-R)in spermatozoa of HBV Infected male patients and the HBV-negative men. Methods: The expression levels of ASGP-R in HBV negative group(n=35), single-positive (HBsAg -[-/HBeAg-) group (n=28)and double-positive (HBsAg-[-/HBeAg~) group (n= 16)were detected with ASGP-R monoclonal antibodies by Western blot and flow cytometry. Results: ASGP-R existed in the plasma membrane and the cytoplasm of human sperm. ASGP-R was highly expressed in human sperm membrane and lowly expressed in human sperm cytoplasm. The expres- sion levels of ASGP-R in negative group, single-positive group and double-positive group were increased gradually. There were significantly higher ASGP-R positive rates in single-positive group and double-posi- tive group than that in negative group(P^0.05). Conclusions: There was ASGP-R expression in the plasma membrane and the cytoplasm of human sperm. HBV might invade into the human sperm by ASGP-R which exist in the plasma membrane. HBV might increase the expression levels of ASGP-R by means of some mechanism.
