雷洛昔芬和结合雌激素对去势大鼠血管舒缩功能的影响

摘　　要：目的通过建立绝经大鼠模型,将雷洛昔芬(RLX)与不同剂量结合雌激素(CEE)配伍,观察两药合用对血管舒缩功能的影响。方法选择9～10月龄健康雌性SD大鼠,分为8组:绝经前基线组、绝经前对照组、绝经后基线组、绝经后对照组、RLX组、CEE组、RLX+低量CEE组及RLX+高量CEE组。行双侧卵巢切除术造绝经造模,术后一周开始药物干预,共10周。采集血标本,检测血浆前列环素(PGI_2)和血栓素A_2(TXA_2)水平。结果大鼠去势后血浆PGI_2和TXA_2的水平同步升高,RLX及CEE治疗后PGI_2和TXA_2的水平同步降低,两药合用的效果与单用RLX或单用CEE相似。结论 CEE与RLX合用对去势大鼠的血管舒缩功能的影响与单用CEE或单用RLX相似,没有进一步增加血栓形成的风险。Objective： To observe the effects of raloxifene （RLX） combined with different doses of conjugated equine estrogen （CEE） on vasomotor parameters in ovariectomized rats. Methods： Sixty-four healthy female SD rats aged 9-10 months were ovariectomized, and then treated with RLX, CEE, and RLX combined with different doses of CEE, respectively, for 10 weeks. The levels of plasma prostacyclin （PGI2） and thromboxane A2 （TXA2） were evaluated. Results： The levels of plasma PGI2 and TXA2 were both increased after ovariectomization in rats （P〈 0.05）, and both reduced after the treatment of drugs （P〈0.05）. There was no significant difference in the levels of PGI2 or TXA2 between raloxifene alone and the combination groups （P〉0.05）. Conclusions： Combination of RLX and CEE did not exert additional effect on vasomotor parameters.
