牙龈卟啉单胞菌牙龈蛋白酶K对人牙周膜细胞增殖的影响

摘　　要：目的探讨牙龈卟啉单胞菌牙龈蛋白酶K对人牙周膜细胞增殖的影响。方法组织块法体外培养人牙周膜细胞,不同浓度的牙龈蛋白酶K作用于人牙周膜细胞24h,对照组为无牙龈蛋白酶K作用的人牙周膜细胞。浓度为12.5μg/ml牙龈蛋白酶K分别作用于人牙周膜细胞24、48、72、96h,对照组也培养相同的时间。采用MTT法检测人牙周膜细胞的增殖,比较实验组与对照组人牙周膜细胞增殖的变化,数据采用单因素方差分析及独立样本t检验。结果牙龈蛋白酶K浓度在6.25～50μg/ml时,作用24h,实验组人牙周膜细胞的增殖低于对照组;牙龈蛋白酶K浓度为12.5μg/ml,作用24～96h,实验组人牙周膜细胞的增殖低于对照组,有显著性差异。结论牙龈卟啉单胞菌牙龈蛋白酶K可抑制体外培养的人牙周膜细胞增殖,并呈剂量和时间依赖性。Objective To evaluate the effects of Lys-gingipain（gingipain K） from Porphyromonas gingivalis（P.gingivalis） on the proliferation of human periodontal ligament cells（hPDLCs）.Methods In the experimental group,hPDLCs were treated by gingipain K in different concentrations for 24 hours.In the control group,hPDLCs were treated without gingipain K.hPDLCs were exposed to gingipain K（12.5μg/ml） for 24,48,72 and 96h.Cell proliferation in two groups was determined by MTT method.The data were statistically analyzed.Results When hPDLCs were exposed to 6.25-50μg/ml gingipain K for 24h,hPDLCs proliferation in experimental group was lower than that in control group.When hPDLCs were exposed to 12.5μg/ml gingipain K for 24-96h,hPDLCs proliferation in experimental group was lower than that in control group.Conclusion Gingipain K can inhibit the proliferation of hPDLCs in vitro in dose and time dependent manner.
