摘　　要：研究穿心莲内酯对人卵巢癌SKOV3细胞株生长、凋亡及HER2蛋白表达的影响。方法：采用MTT法检测细胞增殖抑制率；Annex,inv／PI双染流式细胞仪检测穿心莲内酯诱导SKOV3细胞凋亡的凋亡率；Rh123染色荧光显微镜检测细胞线粒体膜电位（MMP）变化；免疫细胞化学法测定穿心莲内酯处理前后SKOV3细胞磷酸化HER2蛋白表达变化。结果：穿心莲内酯对SKOV3细胞生长增殖具有明显抑制作用；作用细胞16h出现MMP下降；可下调磷酸化HER2蛋白表达。结论：穿心莲内酯可明显抑制SK0v3细胞增殖，并诱导其凋亡；其诱导SKOV3细胞凋亡可能与MMP下降、磷酸化HER2蛋白表诀下调有关。To study the influence of andrographolide （AD） on growth and apoptosis of human ovarian cancer SK- OV3 cells and HER2 protein expression. Methods： The cell proliferation rate was tested by MTT assay; SKOV3 cells apop- tosis rate induced by AD was evaluated by annexin V/PI double staining flow cytometry; mitochondria membrane potential （MMP） changes were detected by Rhodamine 123 （Rh123） staining fluorescence microscopy. Phosphorylated HER2 protein expression changes were analyzed by immunocytochemistry before and after AD induced cells. Results： AD significantly in- hibited SKOV3 cells proliferation; MMP declined when cells were affected for 16 h and phosphorylated HER2 protein ex- pression reduced. Conclusion： AD can significantly inhibit SKOV3 cells proliferation and induce cells apoptosis; SKOV3 cells apoptosis induced by AD may be associated with MMP declining and down-regulation of phosphorylation of HER2 protein.
