摘 要:目的:了解哈尔滨市手足口病病原分布情况及基因特征,为哈尔滨市手足口病的预防控制工作提供参考。方法:收集哈尔滨市哨点医院的手足口病例样本,采用RT-PCR或real-time PCR方法进行病原核酸检测,病毒核酸阳性标本进行RD细胞培养分离病毒,将分离到的EV71采用RT-PCR的方法进行VP1区全基因扩增并测序,应用DNA Man和DNA Star软件序列整理分析。结果:2008年156份临床标本中EV71 59份,CA16 4份;2009年625份临床标本中EV71146份,CA16 82份,未分型别肠道病毒4份;2010年725份临床标本中EV71 202份,CA16 13份,未分型别肠道病毒36份。结论:通过基因比对2008年-2010年哈尔滨市EV71病毒感染手足口流行型别是EV71 C4亚型,未发现变异。Objective:To investigate the pathogens distribution and gene characteristics in HFMD, and provide the scientific basis for making relevant prevention and control strategy. Methods: The DNA in samples of hand - foot - mouth disease patients collected from designated hospitals were tested by RT - PCR or real - time PCR, and the positive samples were cultured on RD cell for virus isolation. Additionally, VP1 region of EV71 was amplified by RT - PCR and sequenced, and then the sequences of VP1 were analysed using DNA Man and DNA Star. Results: In 2008, there were 59 cases of EV71 and 4 cases of CA16 in total 156 samples. In 2009, there were 146 cases of EV71, 82 cases of CA16 and 4 cases of other EV in total 625 samples.. In 2010, there were 202 cases of EV71, 13 cases of CA16 and 36 cases of other EV in total 725 samples. Conclusion: The results indica- ted the EV71 C4 subgenotype was the major pathogen for the epidemic of hand - foot - mouth disease in the sentinel hospitals of Harbin city from 2008 to 2010, and there was no mutation found.
