摘  要:目的 研究利福平对人SaoS-2 成骨样细胞增殖及乳酸(LD)、乳酸脱氢酶(LDH)和DNA 合成的影响.方法 对人SaoS-2 成骨样细胞株常规培养,分别加入不同浓度(50、250、500、750、1 000μg / ml)的利福平,然后用四唑盐(MTT)法测定24、48h 细胞增殖率,试剂盒比色法测定24h 细胞外液的LD 和LDH 含量,ELISA 法测定细胞5- 溴脱氧尿嘧啶核苷(BrdU)含量.结果 不同浓度利福平均可抑制细胞增殖,且呈剂量±赖性;并使细胞外液LD 和LDH 含量增加,细胞DNA 合成量降低.结论 高浓度利福平可以明显抑制细胞的增殖和DNA 合成并影响细胞的代谢能力,用于骨与关节结核治疗的局部用药时应注意控制药物浓度.Objective To investigate the effects of rifampicin on the proliferation and metabolism of human osteoblast-like cell line SaoS-2 in vitro. Methods Human osteoblast-like SaoS-2 cells were cultured with different concentrations(50, 250, 500, 750 and 1 000 tJ g/ml)of rifampicin. The effects of rifampicin on cell proliferation were detected by methyl thiazolyl tetrazolium (Ml-r) assay after 24h and 48h of treatment; the lactic acid (LA) and lactate dehydrogenase (LDH) contents in culture supernatant were measured by commercial assay kits; cell 5-bromodeoxyuridine (BrdU) intake was measured by ELISA kits. Results SaoS-2 cell proliferation was inhibited by rifampicin in a dose-dependent manner, rifampicin increased content; of LA and LDH in extraeellular fluid and reduced the amount of cellular DNA synthesis. Conclusion High concentrations of rifampicin can inhibit cell proliferation and DNA synthesis, affect cell metabolism of human osteoblast-like SaoS-2 cells in vitro, which suggest that caution should be taken with the drug dosage when rifampicin was used to treat bone and joint tuberculosis.
