摘　　要：目的研究半边旗提取物5F对人鼻咽癌CNE-2Z细胞的抑制作用及可能机制。方法以台盼蓝活细胞计数法测定生长曲线;用MTT比色法检测给药后鼻咽癌细胞的增殖抑制情况;采用FCM检测5F对细胞周期和凋亡的影响;用PI/Hoechst33258荧光双染法和DNA ladder检测凋亡。结果 5F作用后,生长曲线测定和MTT比色法实验结果显示,5F对CNE-2Z细胞的增殖和生长有显著的抑制作用并呈明显的时间-剂量依赖关系;流式细胞仪、DNA ladder和荧光双染法结果提示5F可诱导CNE-2Z细胞凋亡。结论半边旗提取物5F能抑制CNE-2Z细胞的增殖效应,其机制可能与诱导细胞凋亡和引起细胞周期阻滞于G2/M期有关。Objective To study the inhibitory effect and its mechanism of 5F. from Pteris semipinnata L. on human nasopharynge- al carcinoma CNE -2Z cells. Methods The growth curve was drawn by means of counting cell number. MTr assay was adopted to determine the inhibition rate on CNE -2Z cells. The cell apoptosis was detected by flow cytomertic analysis, DNA ladder and Hoechst3358/PI fluorescent staining. Results The result of growth curve and MTI＂ showed that the 5F had a remarkable inhibition on the proliferation and growth of CNE - 2Z cells,in time and does - dependent manner. FCM, DNA ladder and fluorescent staining could detect the cell apoptosis. Conclusion 5F can inhibit the proliferation of CNE -2Z cells and the possible mechanism may be related to inducing the cell apoptosis and arresting the cell cycle in G2/M phase.
