摘 要:目的研究泽泻汤总提物及其两个部位(100%乙醇和50%乙醇部位)对高脂血症大鼠的预防性降血脂及抗氧化作用,并确定有效部位。方法采用高脂模型-预防性给受试物法,即喂饲大鼠高脂饲料的同时灌胃泽泻汤总提物及其两个部位,连续30d,测定大鼠血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白-胆固醇(HDL-C)和丙二醛(MDA)的含量以及超氧化物歧化酶(SOD)的活力,计算脏器指数、低密度脂蛋白胆固醇(LDL-C)、动脉硬化指数(AI)和血清HDL/TC比值,并观察肝脏病理切片。结果泽泻汤总提物及其两个部位均能显著降低高脂血症模型大鼠血清中TC、TG、LDL-C含量及AI值(P<0.05或P<0.01),明显升高血清HDL/TC比值、HDL-C含量(P<0.05或P<0.01),其中100%乙醇部位降低TC作用最强;只有泽泻汤总提物和100%部位能显著升高大鼠血清SOD活力、显著降低MDA含量(P<0.05或P<0.01),其中100%乙醇部位降低MDA含量作用强于总提物。结论泽泻汤及其两个部位均有预防性降血脂作用,100%乙醇部位降低TC和MDA作用最强。抗氧化作用的有效部位为泽泻汤总提物和100%乙醇部位。Objective To investigate the effect of Alismatis Rhizoma Decoction (ARD) and its two fractions (100% alcohol and 50% aqueous alcohol) on the levels of serum lipids and its antioxidant capacity in rats, and define the bioactive fraction of ARD. Methods The model of metabolic disturbance of lipids was used in the present study. The rats were divided into six groups : control, high fat diet ( HFD), positive HFD control, ARD treatment, 100% ethanol fraction treatment and 50% ethanol fraction treatment groups. The rats were treated with different samples once a day for 30d. Serum levels of TC, TG, HDL - C, MDA and the activity of SOD were determined. Results In comparison with HFD control group, the three treatment groups significantly re- duced TC ,TG, LDL - C and the arteriosclerosis index(AI) ( P 〈 0.05 or P 〈 0.01 ) , increased HDL - C/TC and HDL - C ( P 〈0.05 or P〈 0.01). The 100% ethanol fraction had more effect on TC than ARD did and its 50% ethanol fraction. ARD and its 100% ethanol fraction significantly reduced level of serum MDA, increased activity of SOD ( P 〈 0.05 or P 〈 0.01 ). The 100% ethanol fraction had more effect on MDA than ARD did. Conclusion ARD and its 100% alcohol and 50% aqueous alcohol fraction can decrease the serum lipids of hyperlipidemic rats, and 100% ethanol fraction is the most effective on TC and MDA. Only ARD and its 100% ethanol fraction can improve the antioxidant capacity of obese rats.
