摘  要:目的探讨抗CD3抗CEA双特异性单链抗体诱导杀伤细胞(CIK细胞)在体内杀伤胃癌的作用。方法将BGC823细胞种植裸鼠后,建立裸鼠胃癌模型,分为3组,分别经尾静脉注入生理盐水、CIK细胞和CIK细胞+双特异性单抗,每3天治疗1次,共6次,取出肿瘤组织称重,并计算出各组的抑瘤率。结果在体内,CIK组和CIK+双特异性单抗组均可抑制肿瘤生长,抑瘤率分别为27.4%和41.7%,与对照组比较,差异均有统计学意义(均P<0.05)。CIK组与CIK+双特异性单抗组的瘤体积、瘤重、抑瘤率比较,差异均有统计学意义(均P<0.05)。结论 CIK细胞本身可以抑制肿瘤细胞的生长,在加入抗CD3抗CEA双特异性单链抗体后,抑瘤作用明显增强。Objective To investigate the killing effect on gastric cancer cells of CIK cells mediated by bispecific single-chain antibody in vivo. Methods To establish gastric cancer model in nude mice with BGC823 cells. And the mice are divided into 3 groups, which are injected with normal saline, CIK ceils and CIK cells + bispecific antibody, respectively, once every 3 days for 6 times. Then remove and weigh the tumors, and calculate the inhibition rate in each group. Results The CIK cells and CIK cells with bispecif- ic single-chain antibody can both inhibit tumor growth in vivo. To compare with the control group, the inhibi- tion rates were 27.4% and 41.7% respectively (P 〈0. 05). To compare with CIK cells, CIK cells with bis- pecific single-chain antibody can inhibit tumor growth (P 〈 0. 05). Conclusion CIK cells alone can inhibit tumor cell growth, associated with the anti-CEA anti-CD3 bispecific single-chain antibody can significantly enhance antitumor effect.
