摘 要:目的探讨端粒酶活性与宫颈癌及子宫颈上皮内瘤样变(CIN)的相关性。方法对50例浸润型宫颈癌,152例CIN(CINⅢ级58例、CINⅡ级44例、CIN级Ⅰ50例)和66例正常者的宫颈新鲜组织,用端粒酶重复扩增法-聚合酶链反应(TRAP-PCR)检测端粒酶活性。结果宫颈癌组中,端粒酶阳性42例(84.0%);CINⅢ级中端粒酶阳性33例(56.9%);CINⅡ级中,端粒酶阳性15例(34.1%);CINⅠ级中,端粒酶阳性12例(24.0%)。而66例正常宫颈组织仅有3例(4.55%)端粒酶活性表达,宫颈癌和CINⅢ组织端粒酶活性表达差异有统计学意义(P<0.01),CINⅢ级与正常对照组织端粒酶活性表达差异有统计学意义(P<0.01)。结论端粒酶活性在宫颈癌发生中可能起到重要作用,在子宫颈病变中检测端粒酶活性,对探讨宫颈病变的进展和宫颈癌的发生具有重要意义。Objective To study the relationship between telomerase activity and cervical cancer and cervical intraepithelial neoplasia. Method 50 cases infiltrating cervical cancer, 152 cases cervical intraepi- thelial neoplasia (58 cases CIN Ⅲ ,44 cases CIN 11,50 cases CIN Ⅰ )and 66 cases normal control cervical fresh tissue were measured for telomerase activity using TRAP-PCR. Result We detected strong telomerase activity in 42 cervical cancer(84. 0% )and telomerase was negative in 8 cases;Telomerase was positive in 33 cases CINm (56.9%) , and telomerase was negative in 25 cases; Telomerase was positive in 15 cases CIN Ⅱ (34. 1% ), and telomerase was negative in 29 cases; Telomerase was positive in 12 eases CIN Ⅰ ( 24.0% ) , and telomerase was negative in 38 cases ; Only 3 cases of 66 normal control tissue was positive of telomerase activity( P 〈 0.01 )were significantly greater in cervical cancer and precancerous lesions than in benign lesions and normal control. Conclusion Telomerase activity may play an important role during cervical carcinogenesis. It is asignal for discuss cervical disease advance and cervical carcinogenesis.
