顺铂联合紫花牡荆素抑制人卵巢癌细胞增殖的研究

摘　　要：目的:探讨顺铂(DDP)是否具有敏化紫花牡荆素(CAS)抑制体外培养人卵巢癌SKOV3细胞生长增殖的作用。方法:体外培养SKOV3细胞;用MTT和平皿克隆法检测顺铂、CAS在亚细胞毒性浓度下,以及两者联合对SKOV3细胞生长增殖的影响;用FCM检测CAS对SKOV3细胞周期的影响;Western blot分析p21和cyclin B1蛋白表达的变化。结果:顺铂具有敏化CAS抑制人卵巢癌SKOV3细胞增殖活性的作用;与亚细胞毒性浓度的顺铂和CAS相比,顺铂和CAS联合作用SKOV3细胞集落形成率明显下降(P<0.01);经亚细胞毒性浓度的顺铂和CAS联合作用48h后SKOV3被阻滞于G2/M期,同时cyclin B1蛋白表达降低,p21蛋白表达增高。结论:顺铂具有敏化CAS抑制体外培养人卵巢癌SKOV3细胞生长增殖的作用,其机制可能是通过降低cyclin B1表达、活化p21实现的。Objective：To investigate whether proliferation of human ovarian cancer cells can be inhibited by combination of DDP and casticin（CAS） in vitro. Methods：Human o- varian SKOV3 cells were cultured in vitro. The inhibitory effect of CAS and DDP at sub-toxic concentration on the viability of human ovarian cancer SKOV3 cells was evaluated by the MTT assay. The phenotypes of colony formation were detected by plate clone formation assay. Phase distribution of cell cycle was analyzed by FCM. The expression of proteins related to prolifera- tion was analyzed by Western blot. Results：The viability of human ovarian cancer SKOV3 cells was significantly inhibited by combination of CAS and DDP at sub-toxic concentration in time- dependent manner. The colony-forming rate was reduced dramatically compared with NS group （ P〈0.01 ） ： The cells treated with combination of CAS and DDP at sub-toxic concentration were remarkably accumulated in G2/M phase by FCM analysis. Western blot showed that expression of p21 protein was up-regulated and protein level of cyclin B1 was depressed after treatment with combination of CAS and DDP at sub-toxic concentration at 48h. Conclusion：DDP at sub- toxic concentration sensitizes induction of proliferation inhibition by CAS in human ovarian cancer SKOV3 cells in vitro through down-regulation of cyclin B1 expression and activation of p21.
