摘  要:目的:探讨TAp63γ的表达对宫颈癌细胞端粒酶活性及生长动力学的影响。方法:通过瞬时转染TAp63γ真核表达载体提高SiHa细胞中TAp63γ的表达水平并经RT-PCR及Western blot验证;通过PCR-ELISA试剂盒检测细胞的端粒酶活性,通过MTT检测细胞增殖及对顺铂的敏感性,通过衰老相关β-半乳糖苷酶染色法检测衰老细胞。结果:与对照组相比,转染TAp63γ的SiHa细胞中端粒酶活性显著下降,差异有统计学意义(P<0.05);细胞增殖减慢,差异有统计学意义(P<0.05);对顺铂敏感性增强,差异有统计学意义(P<0.05);对照组细胞衰老比例为(8±1.5)%;而转染TAp63γ组的衰老细胞比例为(39±8.6)%,明显高于对照组,差异有统计学意义(P<0.05)。结论:TAp63γ具有抑制宫颈癌细胞端粒酶活性的作用,可能是针对宫颈癌细胞生长动力学治疗的新靶点基因。tivity and growth d Objective:To explore the influence of TAp63γ expression on telomerase acynamics of cervical cancer cells. Methods:The expression of TAp63γ was detected by RT-PCR and Western blot after transient transfection of TAp63γ eukaryotic expres- sion plasmid. Telomerase activity was measured by PCR-ELISA kits;cell proliferation and drug sensitivity to cisplatin were detected by MTT;senescence cells were assayed by senescence β- Galactosidase staining kit. Results:Compared with control cells, the telomerase activity of SiHa cells transfacted TAp63γ was decreased sharply with statistic significance(P〈0.05 ). Cell pro- liferation of SiHa cells transfacted TAp63γ was slower than that in control cells (P〈0.05). Drug sensitivity of SiHa cells transfacted TAp63γ to cisplatin was higher than that in control cells(P〈0.05). Proportion of senescence cells of SiHa cells transfacted TAp63γ was (39.0+ 8.6) %, while that in control cells was ( 8.0± 1.5 ) %, with statistic significance ( P 〈0.05 ). Conclusions :TAp63γ can inhibit telomerase activity of cervical cancer cells, and may be a new target gene against growth dynamics of cervical cancer cells.
