摘 要:目的研究IFN-α-2b对K562细胞黏附功能及黏着斑激酶(FAK)mRNA表达的影响。方法 1.采用Cyto Tox96RNon-Radioactive Cytotoxicity Assay Kit试剂盒检测IFN-α-2b对K562细胞黏附功能的影响。2.流式细胞术检测IFN-α-2b作用于K562细胞前后整合素β1表达水平的变化。3.应用RT-PCR技术检测不同浓度IFN-α-2b对K562细胞FAK mRNA表达水平的影响。结果 1.K562细胞高表达整合素β1。IFN-α-2b可提高K562细胞与纤维黏连蛋白的黏附率,并且可降低整合素β1的表达水平。2.当IFN-α-2b的水平为250万~1 000万U.L-1时,随IFN-α-2b水平的增加FAK mRNA的表达逐渐增高。结论 K562细胞中可能存在整合素β1活化障碍。IFN-α-2b可能是通过增加K562细胞的黏附功能反馈性降低整合素β1的表达水平。IFN-α-2b可能通过增加正常FAK mRNA表达水平来改善β1介导的黏附信号通路的缺陷。To investigate the effects of interferon(IFN) -a- 2b on adhesion of K562 cells to fibronectin, and the expres- sion of focal adhesion kinase mRNA of K562 ceils. Methods 1. The effects of IFN - α - 2b on the adhesion of K562 cells to fibronectin were evaluated by Cyto Tox96R Non - Radioactive Cytotoxicity Assay. 2. The effects of IFN - α - 2b on the expression level of integrin β1 on the surface of K562 cells were evaluated by flow cytometry. 3. The expression levels of focal adhesion kinase mRNA were detected by reverse tran- scritptase - polymerase chain reaction of K562 cells treated with different concentrations of IFN - α - 2b. Results 1. Integrin βl expression on the surface of K562 cells was significantly higher. IFN - c~ - 2b could increase the ratio of adhesion of K562 cells to fibronectin, and it could reduce the expression of integrin 13.2. The expression of FAK mRNA increased gradually when the K562 cells were treated with different concentrations of IFN - c~ - 2b, within 2 500 000 to 10 000 000 U . L -1. Conclusions Integrin 131 may be inactive. IFN - α - 2b can reduce the expression of integrin 131 by improving the ratio of adhesion of K562 cells to fibronectin. IFN - α - 2b can up - regulate the normal expres- sion of focal adhesion kinase of K562 cell,and can regain integrin 131 mediated negative proliferation regulation .
