摘 要:目的了解包含WW域的氧化还原酶(WWOX)基因在儿童白血病中的表达,分析WWOX基因与儿童白血病的关系;探讨WWOX基因在儿童白血病中的发病机制。方法收集湖南省人民医院和长沙市其他医院2009年1月-2011年3月白血病患儿51例作为白血病组,同期非白血病患儿10例作为对照组,本研究经患儿监护人知情同意,采用巢式-聚合酶链反应检测2组儿童骨髓单个核细胞中WWOX mRNA表达,应用SPSS 17.0统计软件进行数据分析。结果白血病组患儿WWOX mRNA表达阳性率明显低于对照组(31.37%vs 100.00%,P<0.01);白血病缓解组患儿WWOX mRNA表达阳性率亦明显低于对照组(33.33%vs100.00%,P<0.01),2组比较差异有统计学意义;白血病组患儿WWOX mRNA表达阳性率在不同性别、年龄、白血病类型、治疗阶段、肿瘤负荷组间比较差异均无统计学意义(Pa>0.05)。结论儿童白血病患儿中WWOX mRNA的表达阳性率较低;WWOX mR-NA的表达阳性率可能与儿童白血病的发生发展有关。Objective To investigate the expression of WW domain - containing oxidoreductase (WWOX) in childhood leukemia, and to analyze the relationship between WWOX and childhood leukemia, and to explore the role of WWOX in the pathogenesis of childhood leuke- mia. Methods Fifty - one cases of children with leukemia from Jan. 2009 to Mar. 2011 were collected as the leukemia group, 10 cases of children without leukemia were collected from the People's Hospital of Hunan Province and other hospitals in Changsha during the same peri- od as the control group with the guardians'informed consent, the expression of WWOX mRNA was detected from the bone marrow mononucle- ar cells (BMMCs) of 2 groups using nested - polymerase chain reaction. The data were analyzed by SPSS 17.0 statistical software. Results The positive rate of WWOX mRNA in the leukemia group was significantly less than that of the control group (31.37% vs 100.00% ,P 〈 0.01 ) ; The positive rate of WWOX mRNA in the remission group was significantly, less than that of the control group ( 33.33% vs 100.00% , P 〈0.01 ). In the leukemia group, no significant difference were found in the positive rate of WWOX mRNA between groups differ from age, sex, leukemia type, treatment phase and tumor load ( Pa 〉 0, 05 ). Conclusions Children with leukemia had a low expression of WWOX mRNA; There was no significant difference in the positive rate of WWOX mRNA among leukemia children differ from sex, leukemia type, treatment phase and tumor load.
