灯盏花素对D-半乳糖小鼠生殖器官的影响

摘　　要：目的:研究灯盏花素(Breviscapine,Bre)对D-半乳糖引起衰老小鼠生殖器官异常改变的影响。方法:雌性昆明种小鼠皮下注射D-半乳糖(120 mg/kg.d)67天建立衰老模型,在皮下注射D-半乳糖41天开始,药物组小鼠腹腔注射灯盏花素5、10和20 mg/kg.d,共27天,在皮下注射D-半乳糖第67天记录小鼠体重,断颈处死小鼠,取子宫、卵巢称重,计算器官指数,子宫、卵巢4%甲醛固定HE染色,观察其形态学变化。结果:D-半乳糖明显引起小鼠体重增加,子宫、卵巢指数显著降低,子宫、卵巢组织结构发生异常改变。与空白对照组相比,D-半乳糖组小鼠体重增加了51.95%(P<0.01),卵巢和子宫指数分别降低了42.86%和44.12%。5、10和20 mg/kd.d灯盏花素给药67天不同程度地降低D-半乳糖引起小鼠的体重增加、卵巢和子宫指数的降低及卵巢和子宫组织结构异常改变。结论:灯盏花素对D-半乳糖引起小鼠体重增加,生殖器官萎缩、组织结构异常具有一定的改善作用。Objective： To study the effect of Breviscapine on abnormal change of reproductive organs of aging mice induced by D - galactose. Methods： The models of aging mice were established by subcutaneous injection of D - galactose （ 120 mg/kg^-1 · d^-1 ） among female mice for 67 days, from the 41th day after subcutaneous injection of D-galactose, the mice in drug group were treated with intraperitoneal injection of different dosages of Breviscapine （5, 10, and 20 mg/kg^-1 · d^-1 ） for 27 days; from the 67th day after subcutaneous injection of D - galaetose, the body weight of the mice was recorded, then the mice were killed, their uteri and ovaries were obtained for weighting, the organ index was calculated, then the samples of uteri and ovaries were treated with 4% formaldehyde fixation and HE staining, the morphological changes were observed. Results： D - galactose could increase the body weight of the mice, reduce the uterine index and ovarian index, the abnormal changes of uterine and ovarian tissues and structures were found. Compared with blank control group, the body weight of the mice in D - galactose group increased by 51.95% （P 〈0. 01 ） ; the ovarian index and uterine index reduced by 42. 86% and 44. 12%, respectively. After treating the mice with different dosages of Breviseapine （5, 10, and 20 mg/kg^-1 · d^-1 ） for 67 days, the increase of body weight, the decreases of ovarian index and uterine index, and the abnormal changes of ovarian and uterine tissues and structures of mice induced by D - galactose was prohibited with varying degrees. Conclusion： Breviseapine has a certain improving effect on the increase of body weight, atrophy of reproductive organs, and abnormalities of ovarian and uterine tissues and structures induced by D -galactose.
