酸性磷酸酶在宫颈癌发生过程中的表达及意义

摘　　要：目的研究酸性磷酸酶(ACP)在宫颈癌发生过程中的表达及其临床意义。方法采用RT-PCR、Western blot和免疫组化法分别检测30例正常宫颈,22例宫颈上皮内瘤变(CIN)Ⅰ～Ⅱ级,22例CINⅢ级以及30例宫颈鳞状细胞癌患者组织中ACP的表达情况。结果 RT-PCR结果显示病变宫颈组织较正常宫颈组织相比,ACPmRNA水平的表达呈增高趋势;Western blot和免疫组化结果显示ACP在宫颈癌发生过程中的各级别病变中呈明显增高趋势且差异有统计学意义,同时提示ACP主要定位于细胞浆。结论 ACP在宫颈癌发展过程中表达量明显升高,可作为宫颈癌及癌前病变的一项辅助诊断指标。Objective To investigate the expression and clinical significance of acid phosphatase（ACP） in the occurrence of cervical carcinoma.Methods RT-PCR,Western blot and immunohistochemical method were used to detect the expression levels of ACP in four groups： 30 cases of normal cervical tissues,22 cases of CINⅠ～Ⅱ,22 cases of CINⅢ and 30 cases of cervical squamous cell carcinomas.Results RT-PCR results showed that ACP was higher expressed in diseased cervical tissues than that in normal cervix.Immunohistochemical results indicated the expression of ACP was increasingly higher with the progress of the cervical lesions,as well as the result of Western blot.Simultaneously,it was demonstrated that ACP protein localized in cytoplasm.Conclusion The expression of ACP in the progress of the cervical lesions is increasing remarkably,and ACP may be considered as a auxiliary diagnosis index of CIN and cervical squamous cell carcinomas.
