摘  要:目的探讨三阴性乳腺癌(TNBC)中细胞周期检查点激酶1(Chk1)和多肿瘤抑制基因(p16)的表达及其与临床病理特征的关系,并分析Chk1与p16的相关性。方法应用免疫组化法检测47例TNBC组织、39例非三阴性乳腺癌(non-TNBC)组织及26例乳腺癌癌旁组织中Chk1和p16的表达情况。结果 Chk1在TNBC中的阳性表达率为68.09%,在non-TNBC组织中为46.15%,在癌旁组织中为19.23%,三者比较差异有统计学意义(P<0.05)。Chk1的表达与TNBC病理分级有关,与肿瘤大小、淋巴结转移、月经状态、病理类型及临床分期无关。p16在TNBC组织中的阳性表达率为27.66%,在non-TNBC组织中为48.72%,在癌旁组织中为84.62%,三者比较差异有统计学意义(P<0.01)。p16的表达与TNBC病理分级有关,与肿瘤大小、淋巴结转移、月经状态、病理类型及临床分期无关。TNBC组织中Chk1和p16的表达呈负相关(r=-0.393,P<0.05)。结论 Chk1在乳腺癌组织中阳性表达率较高,在TNBC组织中更明显;p16在乳腺癌组织中阳性表达率较低,在TNBC组织中更明显。TNBC组织中Chk1和p16的阳性表达率分别在不同病理分级之间均存在差异性,且两者呈负相关。Objective To investigate the expression of checkpoint kinase 1(Chk1) and multiple tumor suppressor 1(MST-1/p16) in triple negative breast cancer(TNBC)and its relationship with the clinicopathologic features,and to analyze the relevance of Chk1 and p16 in TNBC.Methods Immunohistochemical method was used to detect the expression of Chk1 and p16 in TNBC,non-TNBC and adjacent normal tissues of breast cancer.Results The positive rate of Chk1 was 68.09% in TNBC,46.15% in non-TNBC and 19.23% in adjacent normal tissues with statistical significance(P〈0.05).Its expression was positively correlated with histological grade,but not with tumor size,lymph node metastasis,menstrual status,pathological types and clinical stage.The positive rate of p16 was 27.66% in TNBC,48.72% in non-TNBC and 84.62% in adjacent normal tissues with statistical significance(P0.01).Its expression was correlated with histological grade,but not with tumor size,lymph node metastasis,menstrual status,pathological types and clinical stage.The expression of Chk1 and p16 in TNBC had an inverse correlation(r=-0.393,P〈0.05).Conclusion Chk1 is over-expressed in TNBC and non-TNBC,especially in TNBC;p16 is under-expressed in TNBC and non-TNBC,especially in TNBC.Chk1 and p16 are correlated with histological grade in TNBC and have an inverse correlation.
