摘  要:目的:定量分析PTEN和HAb18G在非小细胞肺癌组织中的表达特点,并探讨其相互关系。方法:应用免疫组化SP法检测肺癌、肺良性病变和正常肺组织中PTEN和HAb18G蛋白的表达,用ImageProPlus图像分析软件对PTEN和HAb18G蛋白的阳性单位(PU)进行定量测试。结果:PTEN蛋白在肺癌组织中的表达明显低于肺良性病变和正常肺组织(P<0.05),与肺癌分化、淋巴结转移及pTNM分期均无关(P>0.05),周围性大于中央型(P<0.05)。HAb18G蛋白在肺癌组织中的表达明显高于肺良性病变和正常肺组织(P<0.05),与肺癌分化、淋巴结转移及pTNM分期均有关(P<0.05)。不同类型的肺组织中PTEN和HAb18G蛋白表达之间呈负相关(r=-0.312,P=0.002)。结论:PTEN的低表达或缺失,可能与HAb18G的异常激活有一定关系,从而对肺癌的发生、发展起重要作用。Objective To investigate the expression of PTEN and HAb18G protein quantiatively and its correlation in non small lung carcinoma. Methods Immunohistochemistry SP methods were performd to detect the expression of PTEN and HAb18G protein in non small lung cancer, benign lung tissues and normal lung tissues, the intensity (positive unit,PU) of PTEN and HAb18G was assessed quantitatively using ImagePro Plus image analysis softwore. Results PTEN positive intensity in lung carcinoma were significantly lower than that in non-lung carcinoma (P 0.05), it was not associated with histological differentiation,lymph node metastases and pTNM stage. PTEN in peripheral types were higher than in central types(P 0.05). HAb18G positive intensity in lung carcinoma was significantly higher than that in control tissues(P 0.05), and it was related to histological differentiation, lymph node metastases and pTNM stage(P 0.05). PTEN was correlated negatively with HAb18G expression(r = -0.312,P = 0.002). Conclusions The decrease or deletion of PTEN expression may be related to the abnormal activation of HAb18G protein, and palys an important role in the initiation and the development of non small lung cancer.
