摘  要:目的:观察表没食子儿茶素没食子酸酯(EGCG)诱导人胶质瘤细胞(U251)凋亡的生物学效应及对survivin蛋白表达的影响。方法:采用MTT法检测不同浓度EGCG对U251细胞增殖抑制作用;吖啶橙/溴化乙啶(AO/EB)荧光双染法观察各组细胞凋亡形态,计算凋亡率;Wester Blot法检测不同剂量EGCG处理48h对survivin蛋白表达的影响。结果:EGCG显著抑制U251细胞生长,呈浓度依赖性;EGCG可以诱导U251细胞凋亡,细胞凋亡率随着药物浓度的增加而逐渐增加。Wester Blot结果显示EGCG抑制Survivin蛋白的表达。结论:EGCG具有诱导U251细胞凋亡的作用,其机制可能与抑制Sur-vivin蛋白的表达相关。Objective: To investigate the effect of epigallocatechin-3-gallate on Induction of apoptosis in glioblastoma cell line U251 and to study EGCG's influence on the expressions of Survivin protein. Methods .. Ce|l proliferation was examined by MTT assay in U251 cells treated with different concentrations of EGCG. The apoptosis of U251 cells was determined by AO/EB fluorescence stain. Western Blot analysis was performed to detect the expression of Survivin protein. Results..EGCG inhibited the proliferation of U251 cells in a dose-dependent manner. AO/EB fluorescence stain data provided evidence that the apoptosis of U251 cells induced by EGCG in a dose-dependent manner. Western Blot analysis showed that the expression of Survivin protein were effectively inhibited after EGCG was used. Conclusion:EGCG posseses antitumor effect in vitro,its mechanism might be associated with blocking Survivin expressions.
