摘 要:目的观察缺血性脑损伤后大鼠缺血皮质区HDAC家族基因的表达变化规律,探讨HDAC对脑缺血后血管新生的可能影响。方法实验分为缺血后1d组和假手术组(n=3),缺血组采用线栓法制作大脑中动脉阻塞模型,于缺血后1d处死大鼠,取缺血皮质区用于实验;假手术组仅暴露动脉。提取各实验组脑缺血皮质区的总RNA,Real time PCR比较各实验组HDAC家族基因的表达水平。结果脑缺血后HDAC1表达显著上调。结论脑缺血后HDAC1表达显著上调,HDAC家族基因可能在促进脑缺血后血管新生过程中起重要作用。Objective To investigate the expression changes of HDAC genes in impaired rats' cortex after cerebral ischemia,explore the possible influence of HDAC genes on angiogenesis.Methods The rats were divided into tow groups: one day post-ischemia group and sham group.Middle cerebral artery occlusion(MCAO) rat models were made by obstructing MCA using a single strand nylon thread.The rats were killed 1 day after ischemia,and the impaired cortex were extracted for examination.The sham group was not operated as control.Total RNA was isolated from ischemic cortex and sham cortex,and real time PCR was used to determine the expression of HDAC genes.Results The expression of HDAC1 was significantly up-regulated in ischemic cortex after ischemia compared with control.Conclusions The expression of HDAC1 was significantly up-regulated in ischemic cortex after ischemia,indicating a potential role in enhancing angiogenesis after brain ischemia.
